A Scoping Review of Vitamin D for Nonskeletal Health: A Framework for Evidence-based Clinical Practice

Heitor O. Santos et al.

Summary: This review examines the literature on the effects of vitamin D supplementation on nonskeletal health and finds that vitamin D deficiency is associated with various nonskeletal diseases. However, supplementation does not necessarily improve major medical outcomes, even when low 25(OH)D levels are treated. Screening for 25(OH)D status remains important, especially for high-risk patients. Overall, vitamin D supplementation should be guided by circulating levels to correct deficiency.

Supplementing Vitamin D in Different Patient Groups to Reduce Deficiency

Pawel Pludowski

Summary: Studies indicate a high prevalence of vitamin D deficiency in both the general population and at-risk groups. Given the association between vitamin D deficiency and various diseases, addressing this concern becomes crucial, especially in situations where routine monitoring is challenging.

Vitamin D, respiratory infections, and chronic disease: Review of meta-analyses and randomized clinical trials

Davaasambuu Ganmaa et al.

Summary: Current evidence from RCTs and meta-analyses of RCTs is inconsistent regarding the effects of vitamin D supplementation on respiratory infections and chronic diseases. Individuals most likely to benefit are those with baseline VDD or with selected high-risk conditions.

Vitamin D and Its Target Genes

Carsten Carlberg

Summary: The vitamin D metabolite 1 alpha,25-dihydroxyvitamin D-3 acts as a ligand for the transcription factor vitamin D receptor (VDR), and regulates the expression of target genes through chromatin changes. VDR binds to genomic DNA and interacts with partner proteins.

Preparation and Evaluation of Low-Dose Calcitriol Dry Powder Inhalation as Host-Directed Adjunct Therapy for Tuberculosis

D. V. Siva Reddy et al.

Summary: This study investigated the efficacy of inhaled calcitriol as a host-directed therapy for tuberculosis. The results showed that the dry powder inhalation of calcitriol significantly improved lung morphology and reduced histopathology in both the lungs and spleens of Mtb-infected mice. However, the inhalation of calcitriol did not induce a significant host antimicrobial response or reduce bacterial burden in the lungs.

Local nebulization of 1α,25(OH)2D3 attenuates LPS-induced acute lung inflammation

Jef Serre et al.

Summary: Local delivery of vitamin D to the lungs efficiently reduced inflammatory cells induced by LPS in bronchoalveolar lavage and slightly attenuated an increase in CXCL5. In cases of severe vitamin D deficiency, despite not significantly minimizing cellular inflammation, local vitamin D prevented epithelial barrier leakage and damage in the lung. Additional research is needed to determine the long-term beneficial effects of local vitamin D delivery on lung inflammation.

Vitamin D and its therapeutic relevance in pulmonary diseases

Shaniya Ahmad et al.

Summary: Vitamin D plays a key role in various cellular processes and has been associated with maintaining bone and calcium homeostasis, as well as regulating host defense, immunity, and inflammation. Studies have shown correlations between vitamin D deficiency and progression of lung disorders, although the underlying mechanism remains unclear. Vitamin D has inhibitory effects on pulmonary inflammation and enhances innate immune defenses through influencing multiple inflammatory cells.
