Advances in surface plasmon resonance-based biosensor technologies for cancer biomarker detection

Ultrasensitive DNA-Biomacromolecule Sensor for the Detection Application of Clinical Cancer Samples

Summary: Diagnostic testing of biological macromolecules is crucial for early warning of diseases and cancer. However, detecting macromolecules with high sensitivity using interface-based sensing methods remains challenging due to limited surface area and steric hindrance. In this study, a biphasic replacement electrochemical aptamer-based (BRE-AB) sensing strategy is introduced, which achieves ultrasensitive detection of luteinizing hormone (LH) by capturing biomacromolecules in a homogeneous solution and replacing them with small diameter single-stranded DNA to attach to the interface. Molecular Dynamics simulations are used to investigate the binding mechanism between the aptamer and target LH. Furthermore, the BRE-AB sensor demonstrates excellent sensing performance in whole blood and undiluted plasma, enabling successful quantification of LH concentrations in clinical samples, and identifying higher expression of LH in breast cancer patients. The sensor is also simple, low-cost, easy to regenerate and reuse, making it potentially applicable for point-of-care diagnostics of biological macromolecules.

Bimodal Multiplexed Detection of Tumor Markers in Non-Small Cell Lung Cancer with Polymer Dot-Based Immunoassay

Summary: Semiconducting polymer nanoparticles (Pdots) are promising probes for use in fluorometric immunochromatographic test strips (ICTS) due to their ultrahigh brightness, minimal nonspecific adsorption, and multicolor availability. A bimodal readout ICTS platform based on gold-Pdot nanohybrids was developed for the detection of abnormal biomarkers in lung cancer patients' blood, providing rapid qualitative screening and advanced quantitative measurements with high detection sensitivity. The accuracy and reliability of this ICTS platform were demonstrated with clinical serum samples, showing good consistency with results from electrochemiluminescence immunoassays.
