Bacterial proton motive force as an unprecedented target to control antimicrobial resistance

Bingqing Yang et al.

Summary: There is an urgent need for novel antibacterial therapies to combat multidrug-resistant pathogens. Identifying new antimicrobial targets is crucial to avoid cross-resistance issues. Bacterial proton motive force (PMF), which regulates various biological processes, can be a potential target for antibacterial agents.

Inhibiting fatty acid synthesis overcomes colistin resistance

Lindsey A. A. Carfrae et al.

Summary: Treating multidrug-resistant infections has become more reliant on last-resort antibiotics, such as polymyxins. However, the prevalence of polymyxin resistance is increasing, leading to higher mortality rates. Plasmid-borne colistin resistance is a global threat that may diminish the effectiveness of polymyxins. Restoring sensitivity to polymyxins by combining them with sensitizing drugs shows promise in reviving their clinical utility.

Melatonin prevents conjugative transfer of plasmid-mediated antibiotic resistance genes by disrupting proton motive force

Yuqian Jia et al.

Summary: Melatonin has been found to effectively inhibit the transmission of antibiotic resistance genes, providing a potential strategy to prevent the spread of antibiotic resistance. It achieves this by reducing bacterial membrane permeability, inhibiting oxidative stress, and disrupting bacterial proton motive force.

Bismuth Drugs Reverse Tet(X)-Conferred Tigecycline Resistance in Gram-Negative Bacteria

Tian Deng et al.

Summary: Antibiotic resistance poses a serious threat to public health and human safety. The emergence of the novel resistance gene tet(X4) and its variants has jeopardized the clinical utility of tigecycline, a last-line antibiotic for multidrug-resistant bacterial infections. However, the combination of bismuth drugs, particularly bismuth nitrate [Bi(NO3)(3)], with tigecycline has shown to effectively enhance its antibacterial activity against tet(X)-positive bacteria. This is achieved by inhibiting the enzymatic activity of the Tet(X) protein through competitive and noncompetitive binding mechanisms.

Felodipine enhances aminoglycosides efficacy against implant infections caused by methicillin-resistant Staphylococcus aureus, persisters and biofilms

Shutao Zhang et al.

Summary: Felodipine has antibacterial effects against MRSA, biofilm, and persisters. It can inhibit the expression of proteins associated with aminoglycoside resistance, eradicate formed biofilms and kill persisters. Felodipine also reduces MRSA virulence and modulates bacterial metabolism. It inhibits the growth of persisters by binding to a protease. In a mouse infection model, felodipine in combination with gentamicin alleviates bacterial burden and inflammation.

A synthetic lipopeptide targeting top-priority multidrug-resistant Gram-negative pathogens

Kade D. Roberts et al.

Summary: The emergence of multidrug-resistant Gram-negative pathogens poses a global medical challenge. Polymyxin antibiotics are often the last therapeutic option against these pathogens, but they have limitations in safety and efficacy. This study reports the discovery and development of a synthetic lipopeptide that shows improved safety and efficacy against top-priority multidrug-resistant Gram-negative pathogens.

Commercialized artemisinin derivatives combined with colistin protect against critical Gram-negative bacterial infection

Yonglin Zhou et al.

Summary: Artemisinin derivatives, such as DHA, can produce a synergistic antibacterial effect with colistin against most Gram-negative bacteria, particularly those carrying the mcr-1 gene. Mechanism analysis reveals that DHA directly inhibits the activity of MCR-1 and affects the flagellar assembly and energy metabolism of bacteria. In mouse infection models, combination therapy shows significant treatment benefits.

A Marine Antibiotic Kills Multidrug-Resistant Bacteria without Detectable High-Level Resistance

Shang Chen et al.

Summary: Equisetin, derived from marine natural products, shows promising antibiotic activity against multidrug-resistant bacteria such as MRSA and VRE, and can also restore sensitivity to antibiotics in colistin-resistant bacteria. Moreover, low-level equisetin-resistant Staphylococcus aureus displays collateral sensitivity to multiple classes of existing antibiotics and decreased capacity to produce biofilm. Additionally, equisetin is effective against MRSA in infected animal models, indicating its potential as a lead compound in overcoming antibiotic resistance and providing new insights for future antibiotic discovery and development.

Diclofenac Resensitizes Methicillin-Resistant Staphylococcus aureus to β-Lactams and Prevents Implant Infections

Shutao Zhang et al.

Summary: The study shows that high-dose diclofenac inhibits the growth of MRSA without inducing drug-resistant mutations easily, while low-dose diclofenac can resensitize bacteria to beta-lactams and inhibit biofilm formation. Transcriptomic and proteomic analyses indicate that diclofenac reduces the expression of genes and proteins associated with antibiotic resistance and biofilm formation. Murine implant infection models suggest that diclofenac combined with beta-lactams can substantially alleviate MRSA infections in vivo, offering promising applications for preventing perioperative infections.
