An intelligent, autocatalytic, DNAzyme biocircuit for amplified imaging of intracellular microRNAs

Summary: DNAzymes hold great promise as transducing agents for the analysis of intracellular biomarkers. However, their low delivery efficiency and limited signal amplification capability hinder their application in low-abundance biomarker analysis. In this study, a general strategy to design an intelligent, autocatalytic DNAzyme biocircuit is developed for amplified microRNA imaging in living cells. The DNAzyme biocircuit is constructed based on a nanodevice composed of catalytic hairpin assembly (CHA) and DNAzyme biocatalytic functional units, sustained by Au nanoparticles (AuNPs) and MnO2 nanosheets (CD/AM nanodevices). The intelligent autocatalytic multilayer DNAzyme biocircuit can effectively avoid signal leakage and obtain high amplification gain, expanding the application of programmable complex DNA nanocircuits in biosensing, nanomaterial assembly, and biomedicine.

Ultrarapid Microwave-Assisted Synthesis of Fluorescent Silver Coordination Polymer Nanoparticles and Its Application in Detecting Alkaline Phosphatase Activity

Summary: Fluorescent silver coordination polymer nanoparticles (Ag-TPA CPNs) were synthesized via an ultrarapid microwave-assisted strategy using terephthalic acid (TPA) and silver nitrate. The Ag-TPA CPNs showed high fluorescent quantum yield and large Stokes shift, and were used for the detection of alkaline phosphatase (ALP) activity. The method displayed a linear relationship between fluorescence intensity and ALP concentration, with high selectivity and accuracy in biosample analysis.

A Strategy for the Determination of Alkaline Phosphatase Based on the Self-Triggered Degradation of Metal-Organic Frameworks by Phosphate

Interactions of Amino Group Functionalized Tetraphenylvinyl and DNA: A Label-Free On-Off-On Fluorescent Aptamer Sensor toward Ampicillin

Summary: This paper investigates the interaction between aggregation-induced emission luminogens (AIEgens) and nucleic acids and discovers the formation of AIE/DNA complex, leading to the quenching of AIE molecules' fluorescence. The quenching process is demonstrated to be static quenching, and electrostatic and hydrophobic interactions are found to promote the binding process. Based on this interaction, a label-free on-off-on fluorescent aptamer sensor is constructed for the detection of ampicillin (AMP), with a linear detection range of 0.2-10 nM and a limit of detection of 0.06 nM. This sensor has been successfully applied to detect AMP in real samples.

Midas Touch: Engineering Activity of Metal-Organic Frameworks via Coordination for Biosensing

Plasmon-induced near-infrared fluorescence enhancement of single-walled carbon nanotubes

Summary: In this study, the plasmonic effects of Ag and Au nanoparticles with different geometries on the fluorescence intensity of single-stranded DNA-wrapped single-walled carbon nanotubes (ssDNA-SWCNTs) were systematically investigated. Chirality-dependent fluorescence enhancement was observed, which varied with nanoparticle shape and material. Au nanorods increased the fluorescence of (7, 5) and (7, 6) chiralities by 80% and 60% respectively, while Ag nano triangles increased the fluorescence of (7, 5) and (6, 5) chiralities by 200% and 240% respectively. Finite element modeling confirmed contributions from a plasmon-induced localized increase in electron density and radiative recombination of dark exciton states from the resulting electromagnetic field.
