A Pathogenic Variant Reclassified to the Pseudogene PMS2P1 in a Patient with Suspected Hereditary Cancer

Veronica Fragoso-Ontiveros et al.

Summary: The PMS2 gene is involved in DNA repair and its pathogenic variants are associated with Lynch Syndrome. We report a case of a breast cancer patient with a pathogenic variant in the PMS2 gene. This variant was also detected in family members and unrelated individuals, suggesting it may not be hereditary.

Rare Hereditary Gynecological Cancer Syndromes

Takafumi Watanabe et al.

Summary: Hereditary cancer syndromes, characterized by onset at an early age and an increased risk of developing certain tumors, are caused by germline pathogenic variants in tumor suppressor genes and inherited in an autosomal dominant manner. Molecular genetic discoveries have changed cancer diagnosis and management, shedding light on important molecular regulatory pathways in tumor development and treatment. Understanding the association between genotypes and phenotypes in one disease provides insights into the etiology of seemingly different diseases.

Double heterozygous pathogenic variants prevalence in a cohort of patients with hereditary breast cancer

Thais Baccili Cury Megid et al.

Summary: Hereditary breast cancer research found that 19.5% of patients with early onset breast cancer and/or family history of cancer carried germline pathogenic variants in high or moderate-penetrance breast cancer genes. The most common variants were found in TP53, BRCA1, and BRCA2 genes. The co-occurrence of multiple germline pathogenic variants in different genes was observed in a small percentage of the population. There were no significant differences in age of breast cancer onset and risk for bilateral breast cancer between carriers with multiple germline variants and carriers with one variant.

Recurrent NOMO1 Gene Deletion Is a Potential Clinical Marker in Early-Onset Colorectal Cancer and Is Involved in the Regulation of Cell Migration

Jesica Perez-Garcia et al.

Summary: The incidence of early-onset colorectal cancer (EOCRC) is increasing globally, and the molecular mechanisms behind it remain unclear. This study reveals that the NOMO1 gene can be inactivated through deletion or mutation in EOCRC tumors. While the loss of NOMO1 may be a passenger mutation in the development of EOCRC, it significantly enhances colon cancer cell migration.

Summary: In this study, pathogenic variants in 25 cancer susceptibility genes in Turkish BC and CRC patients were profiled, revealing that 17.2% of high-risk BC patients and 26.4% of high-risk CRC patients carried pathogenic variants. The majority of these variants were clinically actionable. BRCA1/2 and MLH1, MSH2, MSH6 genes accounted for two-thirds of the detected pathogenic variants in high-risk patients.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
