Piezo1 activation augments sickling propensity and the adhesive properties of sickle red blood cells in a calcium-dependent manner

Elie Nader et al.

Summary: Haemoglobin S polymerization in the red blood cells of individuals with sickle cell anaemia can cause cellular alterations. Piezo1 protein activation and Gardos channel activity decrease sickle red blood cell deformability and increase their propensity to sickle and adhere to laminin. These results suggest that Piezo1 may represent a potential therapeutic target for sickle cell anaemia.

Proteomic Analysis of the Role of the Adenylyl Cyclase-cAMP Pathway in Red Blood Cell Mechanical Responses

Elif Ugurel et al.

Summary: This study investigates the role of the cAMP signaling mechanism in modulating shear-induced red blood cell (RBC) deformability and examines changes in the phosphorylation of the RBC proteome. The inhibition of signaling molecules significantly deteriorates shear-induced RBC deformability. The cAMP/PKA pathway regulates RBC deformability by triggering significant alterations in the phosphorylation state of RBCs.

Recent Advances in Sickle-Cell Disease Therapies: A Review of Voxelotor, Crizanlizumab, and L-glutamine

Michael Migotsky et al.

Summary: Sickle-cell disease is a genetic hemoglobin disorder that has negative impacts on quality of life and life expectancy. In the past five years, three new treatments, including L-glutamine, crizanlizumab, and voxelotor, have been approved. This study evaluated the efficacy, side effects, and prescribing data of these three medications in treating sickle-cell disease, and found that they have potential therapeutic effects.

Vaso-occlusive crisis in sickle cell disease: a vicious cycle of secondary events

Summary: VOC remains the most common reason for presentation in SCD patients, but current treatments to shorten VOC have had disappointing results. Exploration of the reasons for ineffective monotherapy and consideration of combination approaches with multiple agents may be necessary to target complex downstream pathways of VOC.
