Association of Inherited Mutations in DNA Repair Genes with Localized Prostate Cancer

Daniel J. Lee et al.

Summary: A study of 2391 localized prostate cancer patients found that the germline mutation rates of DNA repair genes in an academic biobank are low, especially among individuals of African genetic ancestry. The mutation rates of genes with published evidence of association with prostate cancer only exceeded 2.5% in high-risk, very-high-risk localized, and node-positive patients. These findings highlight the importance of risk stratification in localized prostate cancer patients.

European Association of Urology Guidelines on Upper Urinary Tract Urothelial Carcinoma: 2020 Update

Morgan Roupret et al.

Summary: The updated EAU guidelines on UTUC aim to assist clinicians in evidence-based management, providing recommendations on diagnosis, risk stratification, radical and conservative treatment, and prognostic factors. Due to the rarity of UTUC, there is insufficient data to provide strong recommendations. Recommendations are based on the 2017 TNM classification and emphasize individualized treatment based on proposed risk stratification.

The Clinical Implications and Molecular Mechanism of CX3CL1 Expression in Urothelial Bladder Cancer

Guangliang Jiang et al.

Summary: High expression of CX3CL1 is significantly associated with poor prognosis in bladder cancer, promoting cell proliferation and invasion. Knockdown of CX3CL1 resulted in upregulation of CDH1 and downregulation of ETS1, RAF1, and EIF4E, with inhibition of the ERK/MAPK signaling pathway.

Co-Occurring Alteration of NOTCH and DDR Pathways Serves as Novel Predictor to Efficacious Immunotherapy in NSCLC

Zhimin Zhang et al.

Summary: The co-occurrence of NOTCH and at least two DDR pathways in advanced NSCLC patients has been shown to improve immunotherapy efficacy and prolong progression-free survival, indicating a potential genomic predictor for stratifying patients suitable for immunotherapy in the future clinical practice.

Comparison of Genomic Characterization in Upper Tract Urothelial Carcinoma and Urothelial Carcinoma of the Bladder

Kaiwei Yang et al.

Summary: The study revealed significant genomic differences and carcinogenesis prevalence between UTUC and UCB, with differences in mutational landscape and driver genes. This suggests the necessity for site-specific management of patients with urothelial carcinoma. Mutational signatures could serve as screening tools for clinical differential diagnosis between UTUC and UCB.

Exploring the Impact of Treatment Switching on Overall Survival from the PROfound Study in Homologous Recombination Repair (HRR)-Mutated Metastatic Castration-Resistant Prostate Cancer (mCRPC)

Rachel Evans et al.

Summary: This study examined the impact of treatment switching from control to olaparib in oncology trials on overall survival, comparing different adjustment methods. The Rank Preserving Structural Failure Time Model (RPSFTM) was found to be the most appropriate approach for adjusting the treatment switching, which could lead to underestimation of the survival benefit of olaparib in Cohort A of the PROfound study. Further development and validation of methods to estimate the impact of treatment switching are required.

Copy Number Signatures and Clinical Outcomes in Upper Tract Urothelial Carcinoma

Bao Guan et al.

Summary: This study used copy number signatures to explore the clinical significance of molecular stratification in UTUC, identifying six CN signatures with Sig6high associated with a favorable outcome. The results indicate that CN signature assessment for risk stratification is feasible and provides a basis for clinical studies evaluating therapeutic interventions and prognosis.
