A tool for translating polygenic scores onto the absolute scale using summary statistics

Oliver Pain et al.

Summary: There is a growing interest in the clinical use of polygenic scores, and this study presents a method for converting polygenic scores to the absolute scale for binary and normally distributed phenotypes. The method uses summary statistics and has been validated using data from the UK Biobank sample. This study provides a useful tool for accurately interpreting polygenic scores using only summary statistics.

Associations of sleep with psychological problems and well-being in adolescence: causality or common genetic predispositions?

Marije C. M. Vermeulen et al.

Summary: Short and problematic sleep are associated with worse psychological functioning in adolescents, and there are bidirectional associations between sleep and psychological problems. Changes in sleep problems in one individual of an identical twin pair are linked to increases in internalizing and externalizing problems, highlighting the importance of focusing on sleep quality for prevention and intervention.

Association Between Systemic Inflammation and Individual Symptoms of Depression: A Pooled Analysis of 15 Population-Based Cohort Studies

Summary: Evidence from various studies suggests that the effects of systemic inflammation on depression are symptom-specific rather than generalized. Future trials for anti-inflammatory treatment of depression may benefit from targeting individuals with distinct inflammation-related physical and cognitive symptoms.

Biallelic mutations in the death domain of PIDD1 impair caspase-2 activation and are associated with intellectual disability

Taimoor I. Sheikh et al.

Summary: PIDD1 encodes a protein that acts as a sensor for centrosome numbers and p53 activity, and plays a significant role in DNA damage response; Mutations in PIDD1 may lead to intellectual disabilities, disrupting the Death Domain structure and causing loss of function; Complete loss of PIDD1 in mice model shows tolerance or compensation for the loss of function.

Sleep duration: A review of genome-wide association studies (GWAS) in adults from 2007 to 2020

Victoria Garfield

Summary: Research in human sleep genetics has shown that specific sleep phenotypes are somewhat heritable, and recent genetic studies have revealed causal relationships between sleep duration and important disease outcomes. However, the heritability of these traits may not be as high as previously estimated. Genome-wide association studies have made significant advances in the field of sleep genetics.

Evaluation of polygenic prediction methodology within a reference-standardized framework

Oliver Pain et al.

Summary: The study compares the prediction accuracy of eight leading polygenic scoring methods, finding that LDpred2, lassosum, and PRScs perform best on average for methods that calculate multiple polygenic scores, while PRScs performs best for methods that provide a single polygenic score without requiring a tuning sample. The study provides a comprehensive comparison of polygenic scoring methods to guide future implementation in research and clinical settings.
