Intermittent Fasting Activates AMP-Kinase to Restructure Right Ventricular Lipid Metabolism and Microtubules

Summary: Intermittent fasting (IF) extends life span through pleotropic mechanisms, with adenosine monophosphate-activated protein kinase (AMPK) playing a crucial role. AMPK enhances lipid metabolism and modulates microtubule dynamics. Dysregulation of these pathways leads to right ventricular (RV) failure in pulmonary arterial hypertension patients. In rodent models of pulmonary arterial hypertension, IF activates RV AMPK, restoring mitochondrial and peroxisomal morphology and regulating lipid metabolism proteins. Additionally, IF increases electron transport chain protein levels and activity in the right ventricle. RV function measured by echocardiography and hemodynamics correlates positively with fatty acid oxidation and electron transport chain protein levels. IF also normalizes transverse tubule structure by reducing microtubule density.

Currently, there is no specific pharmacological treatment available for NAFLD due to incomplete understanding of its mechanisms, lack of accurate and inexpensive imaging tools, and inadequate non-invasive biomarkers. NAFLD can lead to metabolic-associated steatohepatitis (NASH), liver fibrosis, and hepatocellular carcinoma.

Remodeling of the gut microbiome during Ramadan-associated intermittent fasting

These changes in microbiome composition returned to baseline after cessation of intermittent feeding, and alterations in Lachnospiraceae concentrations were correlated with changes in physiological parameters provoked by intermittent fasting. The study suggests that the upregulation of butyric acid-producing Lachnospiraceae may play a role in the health benefits associated with intermittent fasting.
