Study of hairy root formation and plant regeneration in Nicotiana tabaccum

Summary: Hairy roots are an effective method for secondary metabolites production, and A13 strain of Agrobacterium rhizogenes is appropriate for hairy roots induction in Nicotiana tabacum. Hairy roots cultured in MS medium show the highest biomass, increased antioxidant enzyme activities and protein content. Additionally, regenerated plants exhibit better tolerance to salinity stress.

An Optimized Tobacco Hairy Root Induction System for Functional Analysis of Nicotine Biosynthesis-Related Genes

Summary: Rhizobium rhizogenes-mediated plant hairy root induction is a convenient method for studying root-specific gene function. In this study, we developed an optimized tobacco hairy root induction system using different R. rhizogenes strains and media. We found that the strain LBA9402 exhibited the highest induction rate and could also deliver foreign genes to hairy roots. Overexpression of the MsSPL12 gene significantly increased nicotine production in transgenic hairy roots. Our results suggest that this system could be used for efficient manipulation of tobacco nicotine biosynthesis.
