Genome-wide analysis of cytochrome P450 genes in Citrus clementina and characterization of a CYP gene encoding flavonoid 3′-hydroxylase

Summary: In this study, a comprehensive genome-wide analysis of the CYP superfamily in Citrus clementina genome was conducted, identifying 301 CYP genes grouped into ten clans and 49 families. The reliability of the phylogenetic relationship was supported by the characteristics of gene structures and motif compositions. The expression patterns of CYP genes in citrus fruit peel during development and in response to UV-B were elucidated through RNA-seq data. Furthermore, a UV-B-induced CYP gene (CitF3 ' H) was characterized as a flavonoid 3'-hydroxylase, providing insights into the biosynthesis of 3'-hydroxylated flavonoids in citrus.

Generation of the transgene-free canker-resistant Citrus sinensis using Cas12a/crRNA ribonucleoprotein in the T0 generation

Summary: In this study, transgene-free canker-resistant Citrus sinensis lines were generated within 10 months through Cas12a/crRNA ribonucleoprotein-mediated editing of the CsLOB1 gene. These lines exhibited resistance to canker disease by suppressing canker symptoms and inhibiting the growth of Xcc. The development of these canker-resistant citrus cultivars provides a sustainable and efficient solution for canker control and presents a transgene-free genome editing strategy.

Optimization of in vitro and ex vitro Agrobacterium rhizogenes-mediated hairy root transformation of soybean for visual screening of transformants using RUBY

Summary: In this study, in vitro and ex vitro Agrobacterium rhizogenes-mediated hairy root transformation (HRT) assays were optimized in soybean using the RUBY reporter. Various parameters were evaluated, and it was found that the in vitro assay was more efficient than the ex vitro assay in terms of the percentage of induction of hairy roots and transformed roots. However, the ex vitro technique was considered faster and less complicated. Machine learning-based modeling was used to define optimal protocols for both assays. This study establishes efficient and reliable hairy root transformation protocols applicable for functional studies in soybean.

Efficient floral dip transformation method using Agrobacterium tumefaciens on Cosmos sulphureus Cav.
