Synergistic Interaction of Piperine and Thymol on Attenuation of the Biofilm Formation, Hyphal Morphogenesis and Phenotypic Switching in Candida albicans

Arumugam Priya et al.

Summary: The incidence of fungal infections has increased due to antifungal resistance. Biofilm formation is a major contributor to infectious diseases and antimicrobial resistance. This study investigated antibiofilm and combinatorial approaches to attenuate biofilm formation and virulence factors expression in Candida. Piperine and thymol, bioactive components of pepper and thyme, showed synergistic effects in reducing Candida adherence, hyphal extension, and morphological transformation. Piperine and thymol can be used synergistically for the treatment of biofilm-associated C. albicans infection.

Anti-biofilm activities of coumarin as quorum sensing inhibitor for Porphyromonas gingivalis

Zhiyan He et al.

Summary: This research focused on studying the effect of coumarin on the formation of Porphyromonas gingivalis biofilm, a key pathogen in periodontitis. The results showed that coumarin inhibited the growth of P. gingivalis and caused damage to cell morphology at concentrations above 400 μM. Sub-MIC concentrations of coumarin did not affect cell surface properties, aggregation, attachment, or early-stage biofilm formation. However, it demonstrated anti-biofilm effects on late-stage biofilms and pre-formed biofilm dispersion. The biofilms treated with coumarin became interspersed, and the expression of biofilm-related genes was downregulated. Coumarin also inhibited AI-2 activity and interacted with the HmuY protein through molecular docking analysis.
