Chemical Labeling and Enrichment of Histone Glyoxal Adducts

Devin M. Ray et al.

Summary: In this study, glyoxal modifications on histones were characterized using quantitative proteomics and a GO-reactive probe derived from aniline. It was found that glycation regulatory proteins can reduce the levels of histone GO adducts. A two-round pull-down method was employed to enrich histone H3 GO glycation and map these adducts to specific chromatin regions.

GATD3A, a mitochondrial deglycase with evolutionary origins from gammaproteobacteria, restricts the formation of advanced glycation end products

Andrew J. Smith et al.

Summary: We identified a mitochondrial protein, GATD3A, with bacterial evolutionary affinities, that functions as a deglycase and restricts the formation of AGEs in mitochondria. GATD3A may play a role in maintaining the integrity of important biomolecules.

DJ-1 is not a deglycase and makes a modest contribution to cellular defense against methylglyoxal damage in neurons

Melissa Conti Mazza et al.

Summary: Human DJ-1 is a cytoprotective protein that defends against oxidative stress and mitochondrial dysfunction, but its role as a deglycase is controversial. Our study suggests that DJ-1 is not a true deglycase and has minor effects in protecting neurons against methylglyoxal toxicity.

Double DJ-1 domain containing Arabidopsis DJ-1D is a robust macromolecule deglycase

Melvin Prasad et al.

Summary: This study reveals that the DJ-1D protein in plants has the ability to repair glycated DNA, RNA, and proteins, providing robust tolerance to endogenous and exogenous stresses. The study also shows that DJ-1D can reduce the formation of reactive carbonyls, thereby suppressing the accumulation of advanced glycated end-products. Furthermore, the study indicates the existence of a new DNA repair pathway in plants.

Glycolate combats massive oxidative stress by restoring redox potential in Caenorhabditis elegans

Veronica Diez et al.

Summary: Diez et al. demonstrate that glycolate suppresses the deleterious effects of peroxide on mitochondrial activity and growth in worms. They find that the conversion of glycolate into glycine and serine helps worms maintain cellular antioxidants following oxidative stress. This study suggests that glycolate serves as a natural antioxidant, improving organismal fitness under stressful conditions.
