Excessive ER-phagy contributes to ochratoxin A-induced apoptosis

Huiqiong Deng et al.

Summary: This study reveals that the nephrotoxic effect of ochratoxin A (OTA) on human proximal tubule epithelial (HK-2) cells is mediated by the activation of endoplasmic reticulum stress (ERS) and excessive ER-phagy, leading to apoptosis. The vicious cycle between excessive ER-phagy and ERS contributes to the promotion of apoptosis.

miR-155-5p/FOXO3a promotes pulmonary fibrosis in rats by mediating NLRP3 inflammasome activation

Ling Wu et al.

Summary: This study found that miR-155-5p promotes pulmonary fibrosis by targeting FOXO3a and activating the NLRP3 inflammasome, leading to the release of IL-113 and IL-18. This research provides a new target for the management of pulmonary fibrosis.

Ochratoxin a induces hepatic fibrosis through TGF-β receptor I/Smad2/3 signaling pathway

Seung A. Chae et al.

Summary: This study investigated the mechanism of OTA-induced liver fibrosis and found that OTA induces epithelial-mesenchymal transition (EMT) and hepatic fibrosis through the TGF-beta RI and Smad2/3 signaling pathway. The results showed that OTA exposure increases the expression of liver fibrosis-related markers and elevates damage indicators. Silencing Smad2/3 and inhibiting TGF-RI can reduce the expression of liver fibrosis-related markers.

CCAAT/Enhancer-Binding Proteins in Fibrosis: Complex Roles Beyond Conventional Understanding

Lexun Wang et al.

Summary: CCAAT/enhancer-binding proteins (C/EBPs) are transcription factors that interact with DNA to regulate gene expression. They play critical roles in fibrosis, and studying C/EBPs could lead to new approaches for treating fibrosis.

TGF-β-induced α-SMA expression is mediated by C/EBPβ acetylation in human alveolar epithelial cells

Hui Ding et al.

Summary: The study confirmed that deacetylation of C/EBP beta in A549 cells successfully ameliorated TGF-beta 1-induced EMT, as shown by reduction in alpha-SMA expression and excessive collagen-I accumulation. This indicates that the EMT and fibrotic effect of TGF-beta 1 depend on acetylated C/EBP beta-mediated regulation of alpha-SMA gene activity.

TGF-β Signaling: From Tissue Fibrosis to Tumor Microenvironment

Jeff Yat-Fai Chung et al.

Summary: TGF-beta signaling plays crucial roles in inflammatory diseases by regulating immunocytes in tissue fibrosis and accelerating tumor development in cancer. The pleiotropic nature of TGF-beta signaling in generating fibrotic TME, containing CAFs and matrix proteins, remains largely unclear, despite recent studies demonstrating its clinical implications in cancer.
