Tuning the microstructure and mechanical properties of lyophilized silk scaffolds by pre-freezing treatment of silk hydrogel and silk solution

Summary: This study investigates the effects of pre-freezing treatments at different temperatures (-20℃, -80℃, or -196℃) on the microstructure, mechanical properties, and secondary structure of silk scaffolds prepared from lyophilization of silk hydrogels and silk solutions. The results show that silk hydrogels produce scaffolds with more nanofibrous structures compared to silk solutions. Only silk hydrogels can produce aligned macro-channels, while both silk solutions and hydrogels pre-frozen with liquid nitrogen can generate nanofibrous scaffolds. Silk hydrogel scaffolds are dominated by β-sheets, while silk solution scaffolds are largely amorphous. These findings are important for controlling the microstructure and mechanical properties of silk scaffolds.

Anisotropic dense collagen hydrogels with two ranges of porosity to mimic the skeletal muscle extracellular matrix

Summary: This study developed a novel 3D in vitro model of muscle extracellular matrix (ECM) that replicated the specific characteristics of the native ECM, including its biochemical composition, stiffness, anisotropy, and porosity. The model was created using unidirectional 3D printing of dense collagen with aligned collagen fibrils, and the space between layers was adjusted to facilitate nutrient and oxygen diffusion. By modifying the gelling conditions, the mechanical properties of the construct matched those of the physiological muscle ECM. The addition of large channels allowed for cell colonization without affecting the physical properties of the hydrogel, and myoblasts embedded in the hydrogel differentiated into multinucleated myotubes.

The fibres were shown to be non-toxic to normal human dermal fibroblast cell lines and have the potential for smart delivery fabrics/dressings.

Surface-fill H2S-releasing silk fibroin hydrogel for brain repair through the repression of neuronal pyroptosis

Summary: In this study, a surface-fill H2S-releasing silk fibroin hydrogel (H2S@SF hydrogel) was developed for the treatment of traumatic brain injury (TBI). The results showed that H2S@SF hydrogel inhibited TBI-induced neuronal pyroptosis and alleviated brain edema and neurodegeneration. It also improved motor dysfunction, anxious behavior, and memory impairment caused by TBI. H2S@SF hydrogel had a significant impact on the long-term prognosis of TBI.

Macroporous chitosan/methoxypoly(ethylene glycol) based cryosponges with unique morphology for tissue engineering applications

Summary: This study investigated the effects of different methoxypolyethylene glycol derivatives on the morphology and physical properties of chemically crosslinked, semi-interpenetrating polymer network chitosan/mPEG blend cryosponges. The cryosponges were capable of delivering bioactives and supporting neuronal cell growth.
