FinnGen provides genetic insights from a well-phenotyped isolated population

Mitja Kurki et al.

Summary: Population isolates, like those in Finland, provide valuable insights for genetic research due to the concentration of deleterious alleles in low-frequency variants. FinnGen aims to study the genome and health data of 500,000 Finnish individuals, enabling the identification of new associations and the exploration of their implications for common diseases. The study identified 30 new associations and 2,733 genome-wide significant associations, highlighting the power of bottlenecked populations in uncovering disease biology.

The Role of Serotonin Neurotransmission in Gastrointestinal Tract and Pharmacotherapy

Tomasz Guzel et al.

Summary: Serotonin is a neurotransmitter that acts in both the central nervous system and peripheral structures, also functioning as a hormone in platelets. It modulates CNS neurons, pain transmission, and platelet aggregation. Serotonin also influences muscle motility in the gut, bronchi, uterus, and vessels. Its synthesis starts with hydroxylation and decarboxylation of tryptophan. 5-HT receptors in the brain and gut are targets for drugs that modify serotonin neurotransmission.

Genetics of irritable bowel syndrome: shifting gear via biobank-scale studies

Michael Camilleri et al.

Summary: Genetic predisposition plays a significant role in the pathophysiology of irritable bowel syndrome (IBS). This review provides a comprehensive overview of genetic research in IBS and discusses new concepts in IBS genetics. While genetic research in IBS has been limited and inconclusive, the availability of large international cohorts and population-based biobanks presents unprecedented opportunities for well-powered studies. This review focuses on recent advances in identifying genes involved in digestion, ion channel function, neurotransmitters, neuronal pathways, and gut motility control.

Charting the Landscape of Genetic Overlap Between Mental Disorders and Related Traits Beyond Genetic Correlation

Guy Hindley et al.

Summary: There is extensive genetic overlap across mental disorders and related traits, with mixed effect directions and few disorder-specific variants. This represents a conceptual advance in our understanding of the landscape of shared genetic architecture across mental disorders, which may inform genetic discovery, biological characterization, nosology, and genetic prediction.

Nociceptor neurons direct goblet cells via a CGRP-RAMP1 axis to drive mucus production and gut barrier protection

Daping Yang et al.

Summary: Neuroepithelial crosstalk plays a critical role in gut physiology, particularly in mediating gut barrier protection. Sensory neurons communicate with intestinal goblet cells to promote mucus secretion and gut protection via the CGRP-Ramp1 pathway. Additionally, commensal microbes can activate nociceptors to regulate CGRP release.

Diminished androgen levels are linked to irritable bowel syndrome and cause bowel dysfunction in mice

Daniella Rastelli et al.

Summary: Functional gastrointestinal disorders (FGIDs) exhibit sex differences in incidence, symptoms, and treatment response. Lower levels of free testosterone were found in adults with irritable bowel syndrome (IBS), and it was inversely correlated with symptom severity. Depletion of gonadal androgens caused a deficit in gastrointestinal transit in mice, which could be rescued by restoring a single androgen hormone. Androgen receptors in a subset of enteric neurons became responsive during puberty and were required for normal colonic motility. These findings establish a role for gonadal androgens in bowel function regulation and link altered androgen levels with a common digestive disorder.

The distribution of common-variant effect sizes

Luke J. O'Connor

Summary: The genetic effect-size distribution of a disease, which includes the number of risk variants, their effect sizes, and sample sizes required to discover them, can be accurately estimated using Fourier Mixture Regression (FMR). FMR estimates that 100,000-1,000,000 cases will be needed to find genome-wide significant SNPs explaining 50% of SNP heritability. In large studies, genome-wide significance becomes more conservative and less stringent thresholds can achieve high true positive rates if confounding is controlled.

Extensive bidirectional genetic overlap between bipolar disorder and cardiovascular disease phenotypes

Linn Rodevand et al.

Summary: The study reveals extensive polygenic overlap between bipolar disorder (BIP) and comorbid cardiovascular diseases (CVD), suggesting shared molecular genetic mechanisms. The shared loci exhibit a pattern of mixed effect directions, indicating variation in genetic susceptibility to CVD among BIP subgroups, which may explain the heterogeneity of CVD comorbidity in BIP patients. Therefore, the findings suggest a focus on targeted lifestyle interventions and personalized pharmacological treatment to reduce CVD comorbidity in BIP patients.
