Summary: White matter hyperintensity (WMH) lesions and changes in adjacent normal-appearing white matter can affect structural brain connectivity measurements. The virtual lesion approach can estimate structural connectivity changes caused by WMH. This study utilized diffusion MRI data from young and older subjects to evaluate the impact of using different datasets for virtual lesion tractography. The results suggest that using normative diffusion MRI data from young subjects is preferable for virtual lesion tractography of WMH.

Spatial-temporal patterns of brain disconnectome in Alzheimer's disease

However, the vulnerability of specific neural connections related to WMHs in AD is not well understood. This study used a computational framework to assess the spatial-temporal patterns of WMH-related structural disconnectivity in AD. The results showed a specific pattern of brain disconnectome that predicted conversion from mild cognitive impairment (MCI) to dementia with high accuracy.
