Summary: A series of compounds inhibiting BRD4 were synthesized and compound 3r (ZLD2218) showed the most potent inhibitory activity against BRD4. It effectively alleviated kidney injury and fibrosis in mice models. Mechanistically, ZLD2218 inhibited BRD4 expression and suppressed fibrotic signaling in the kidneys and cells. These findings suggest that ZLD2218 could be a promising lead compound for the treatment of kidney fibrosis.
