AtMCM10 promotes DNA replication-coupled nucleosome assembly in Arabidopsis

Xinjie Zhao et al.

Summary: MCM10 is essential for DNA replication initiation and elongation in yeasts and animals, but its detailed mechanisms in these processes are not well known. In this study, we identified AtMCM10 gene in Arabidopsis and found that it can directly bind to histone H3-H4 and promote nucleosome assembly in vitro. Loss of both AtMCM10 and CAF-1 is embryo lethal, indicating their indispensable roles in replication-coupled nucleosome assembly.

Coordinated histone variant H2A.Z eviction and H3.3 deposition control plant thermomorphogenesis

Fengyue Zhao et al.

Summary: Plants can adjust their development and morphology in response to temperature changes through a process called thermomorphogenesis. The regulation of gene expression reprogramming in this process involves the eviction of a histone variant H2A.Z by INO80 and the deposition of another histone variant H3.3. These processes are coordinated and play a crucial role in gene activation induced by high temperature.

A REF6-dependent H3K27me3-depleted state facilitates gene activation during germination in Arabidopsis

Jie Pan et al.

Summary: Seed germination is a complex process involving extensive changes in metabolism, gene expression, and cellular identity. This study examines the dynamics and function of histone H3 lysine 27 trimethylation (H3K27me3) during germination in Arabidopsis. The results suggest that H3K27me3 reprogramming occurs with a delay compared to transcriptomic changes and that H3K27 demethylation, mediated by RELATIVE OF EARLY FLOWERING 6 (REF6), is necessary for robust germination and the activation of hormone-related and expansin-coding genes.

The histone H3.1 variant regulates TONSOKU-mediated DNA repair during replication

Hossein Davarinejad et al.

Summary: This study reveals the specific function of H3.1 during replication in plants, demonstrating its interaction with TSK and DNA polymerase theta in the absence of H3 lysine 27 monomethylation catalyzed by ATXR5/ATXR6. It also highlights a common strategy used in multicellular eukaryotes for regulating post-replicative chromatin maturation and the recognition of H3.1 variant by histone monomethyltransferases.

Histone H3.3 deposition in seed is essential for the post-embryonic developmental competence in Arabidopsis

Ting Zhao et al.

Summary: This study reveals the crucial role of Arabidopsis histone variant H3.3 in endowing seeds with post-embryonic developmental potentials and regulating germination and post-embryonic development. H3.3 facilitates chromatin opening at regulatory regions, regulates gene transcriptional regulation during germination, and is associated with gene body DNA methylation and the restriction of chromatin accessibility and cryptic transcription at specific gene ends in seeds.

The transcriptional repressors VAL1 and VAL2 recruit PRC2 for genome-wide Polycomb silencing in Arabidopsis

Liangbing Yuan et al.

Summary: The study reveals that the transcriptional repressors VAL1 and VAL2 recruit PRC2 to target loci in Arabidopsis thaliana. They physically interact with PRC2 catalytic subunits SWN and CLF, colocalize at target loci, and regulate H3K27me3 levels, affecting transcription.

The chromatin remodeler DDM1 prevents transposon mobility through deposition of histone variant H2A.W

Akihisa Osakabe et al.

Summary: The study by Osakabe et al. demonstrates that the chromatin remodeler DDM1 plays a crucial role in silencing transposable elements by mediating H2A.W deposition in Arabidopsis, preventing their mobility. This discovery sheds light on the essential mechanism of chromatin remodelers in regulating TEs and provides new insights into the interaction between TEs and their host in the contexts of evolution and disease.

The histone variant H2A.W and linker histone H1 co-regulate heterochromatin accessibility and DNA methylation

Pierre Bourguet et al.

Summary: The study shows that H2A.W antagonizes H1 deposition in heterochromatin, helps maintain heterochromatin accessibility and DNA methylation levels, and co-regulates heterochromatic features with H1. The null mutant created by CRISPR reveals the importance of H2A.W in regulating heterochromatin accessibility.

Histone variants take center stage in shaping the epigenome

Michael Borg et al.

Summary: The dynamic properties of the nucleosome are crucial to genomic activity, with histone variants playing a pivotal role in modulating nucleosome structure and function. Specifically in plant chromatin landscapes, deposition of replicative H3.1 controls epigenetic inheritance while local replacement of H3.1 with H3.3 marks cells undergoing terminal differentiation. Deposition of specialized H3 variants in specific cell types is emerging as a novel mechanism of selective epigenetic reprogramming during the plant life cycle.
