Summary: GPR35 is a chemoattractant receptor that promotes eosinophil migration to the inflammatory mediator 5-HIAA, thereby facilitating the accumulation of eosinophils in fungal meningitis caused by Cryptococcus neoformans. The deficiency of GPR35 inhibits eosinophil recruitment and fungal growth, while overexpression enhances eosinophil homing to airways and fungal replication. The ligand activity of GPR35 comes mainly from activated platelets and mast cells, and inhibiting serotonin conversion to 5-HIAA or reducing 5-HIAA production can enhance the clearance of Cryptococcus. Therefore, the 5-HIAA-GPR35 axis plays a crucial role in modulating the clearance of a lethal fungal pathogen and has implications for the potential use of serotonin metabolism inhibitors in fungal infection treatment.
