The activity of human enhancers is modulated by the splicing of their associated lncRNAs

Jennifer Yihong Tan et al.

Summary: Pervasive enhancer transcription and splicing of enhancer-associated long noncoding RNAs (elncRNAs) play crucial roles in enhancer activity and gene expression regulation. Our study reveals that the splicing motifs within multi-exonic elncRNAs have evolved under selective constraints during human evolution. Using a genome-wide approach, we provide evidence that elncRNA splicing is associated with chromatin signatures at enhancers and target mRNA expression. Our findings demonstrate the importance of efficient elncRNA splicing in enhancer activity.

Disrupted mossy fiber connections from defective embryonic neurogenesis contribute to SOX11-associated schizophrenia

Xianmixinuer Abulaiti et al.

Summary: Abnormal mossy fiber connections in the hippocampus have been found in patients with schizophrenia, but the genetic basis and its contribution to the development of schizophrenia were unclear. This study demonstrated that a high-risk allele of SNP rs16864067 downregulated the SOX11 gene, leading to abnormal polarity in hippocampal neural progenitor cells derived from schizophrenia patients. A mouse model with SOX11 deficiency showed similar abnormal polarity and hippocampal neurogenesis defects, resulting in disrupted mossy fiber connections and schizophrenia-like behaviors. Activation of hippocampal neurogenesis during embryonic development partially alleviated these disruptions and improved schizophrenia-related behaviors.

ADRAM is an experience-dependent long noncoding RNA that drives fear extinction through a direct interaction with the chaperone protein 14-3-3

Wei Wei et al.

Summary: In this study, RNA capture-seq was used to identify a large population of lncRNAs expressed in the infralimbic prefrontal cortex of adult male mice after fear-related learning. A specific lncRNA called ADRAM was discovered, which acts as both a scaffold and a guide to regulate the expression of memory-associated genes and is required for fear extinction memory.

Activity-regulated synaptic targeting of lncRNA ADEPTR mediates structural plasticity by localizing Sptn1 and AnkB in dendrites

Eddie Grinman et al.

Summary: This study identified a long noncoding RNA named ADEPTR, which is up-regulated and synaptically transported in a cAMP/PKA-dependent manner in hippocampal neurons, independently of its protein-coding host gene. ADEPTR mediates dendritic localization through molecular motor protein Kif2A and binds to actin-scaffolding regulators ankyrin (AnkB) and spectrin (Sptn1) to regulate synaptic structural plasticity.
