MMP9: A Tough Target for Targeted Therapy for Cancer

Katarzyna Augoff et al.

Summary: MMP9, a protease with diverse functions, plays a crucial role in tumorigenesis by regulating cancer cell migration, epithelial-to-mesenchymal transition, immune response, angiogenesis, and tumor microenvironment formation. However, the complicated regulation mechanism and high homology with other MMP family members make it difficult to develop effective and safe MMP9 inhibitors as anticancer drugs. The recent development of blocking antibodies shows promise, but further research is needed to fully understand the mechanisms of MMP9 in tumorigenesis and develop new therapies targeting MMP9.

TGFβ biology in cancer progression and immunotherapy

Rik Derynck et al.

Summary: TGF beta signaling plays a key role in cancer progression by promoting cancer cell invasion, dissemination, stem cell properties, therapeutic resistance, and the generation of an immunosuppressive environment. Inhibiting TGF beta signaling is considered a crucial avenue to enhance the efficacy of cancer immunotherapies. Understanding and targeting TGF beta signaling mechanisms in tumors and their microenvironment may provide valuable insights for cancer treatment.

Mesenchymal Stromal Cells in Neuroblastoma: Exploring Crosstalk and Therapeutic Implications

Caroline Hochheuser et al.

Summary: Neuroblastoma is the second most common solid cancer in childhood, with bone marrow being a common site of distant metastasis. Therapy resistance of residual tumor cells in BM remains a major obstacle for the cure of NB. The role of mesenchymal stromal cells in NB development and progression is not fully understood, with a focus on BM as a metastatic niche.

Therapeutic Implications of TGFβ in Cancer Treatment: A Systematic Review

Veronica Gomez-Gil

Summary: While the importance of TGF beta in cancer development has long been recognized, a successful therapy targeting this cytokine has not been developed yet. Various strategies are being tested to regulate TGF beta expression in cancer treatment, but the dual role of TGF beta and its crosstalk with other signaling pathways may compromise treatment effectiveness.

Copper: An Intracellular Achilles' Heel Allowing the Targeting of Epigenetics, Kinase Pathways, and Cell Metabolism in Cancer Therapeutics

Filip Michniewicz et al.

Summary: Copper plays a crucial role in cancer progression by influencing cell processes, and overcoming drug resistance and relapse may require developing broader molecular targeting strategies. Overcoming these limitations may involve using copper chelating agents to simultaneously target multiple cancer treatment mechanisms.

Tetrathiomolybdate (TM)-associated copper depletion influences collagen remodeling and immune response in the pre-metastatic niche of breast cancer

Ying L. Liu et al.

Summary: The study found that Tetrathiomolybdate promotes survival in breast cancer patients by reducing collagen cross-linking and increasing degradation, as well as decreasing collagen deposition and myeloid-derived suppressor cells while increasing T-cell infiltration in the tumor microenvironment. This discovery indicates the potential application of TM across various types of cancer.
