Stimuli-Responsive Self-Degradable DNA Hydrogels: Design, Synthesis, and Applications

Summary: DNA hydrogels are increasingly important in biomedical and bioanalysis applications. They are highly programmable, multifunctional, and biocompatible, making them effective carriers for drugs and bioactive cargoes. However, the stability of DNA hydrogels hinders rapid release of cargoes. Stimulus-responsive DNA hydrogels have been designed to meet these requirements, switching from gel to solution for precision cargo delivery and bioanalysis. This review summarizes innovative methods for designing stimuli-responsive self-degradable DNA hydrogels and discusses their applications and challenges in the fields of bioanalysis and biomedicine.

Core-shell bioprinting of vascularized in vitro liver sinusoid models

Summary: In this study, a liver sinusoid-like model was developed using coaxial extrusion-based 3D bioprinting. The model consisted of a core compartment with pre-vascular structures and a shell compartment with hepatocytes. Alginate and methylcellulose were used as the shell ink, while collagen-fibrin matrices with gelatin were used as the core ink. The addition of human fresh frozen plasma enhanced the viability and cluster formation of HepG2 cells. The triple culture model promoted cellular interactions and albumin secretion. This core-shell bioprinting approach is a valuable tool for studying cell-cell interactions and developing complex tissue-like models.

Capsule-like DNA Hydrogels with Patterns Formed by Lateral Phase Separation of DNA Nanostructures

Summary: Phase separation is crucial in artificial cell construction. This study demonstrates the lateral liquid-liquid phase separation of DNA nanostructures, resulting in the formation of capsule-like hydrogels. The appearance of different phase separation patterns can be controlled by designing DNA sequences and altering the mixing ratio of nanostructures. The extracted hydrogels maintain their patterns and can be combined with enzymatic reactions.
