Unlocking the power of NOX2: A comprehensive review on its role in immune regulation

Kevin Bode et al.

Summary: Reactive oxygen species (ROS) have diverse functions within cells and organisms, but their potential to damage biological structures has long been recognized. While phagocytes use ROS to defend against pathogens, regulated release of signaling ROS (sROS) modulates immune responses. This release is primarily attributed to the enzyme NOX2 expressed in myeloid cells. A comprehensive understanding of NOX2 activation and regulation is crucial for pharmaceutical interventions of ROS-related diseases. In this review, recent progress regarding immune regulation by NOX2-derived ROS and the consequences of its deregulation on immune disorders are summarized.

High-throughput and high-content bioassay enables tuning of polyester nanoparticles for cellular uptake, endosomal escape, and systemic in vivo delivery of mRNA

Yuan Rui et al.

Summary: Nanoparticle-based mRNA therapeutics show great promise, but cellular internalization and endosomal escape pose challenges for cytosolic delivery. Researchers developed a dual nanoparticle uptake and endosomal disruption assay, which can detect cellular internalization and endosomal disruption. The study found that this assay has higher predictive capacity for mRNA delivery compared to conventional polymer and nanoparticle characteristics.

mRNA Delivery and Storage by Co-Assembling Nanostructures with Designer Oligopeptides

Ruilu Feng et al.

Summary: In this study, we introduce a peptide sequence called pepMAX that can co-assemble with mRNA into nanostructures for efficient transfection of multiple cell lines. The transfection efficiency of pepMAX is comparable to that of Lipofectamine MessengerMAX (TM) in HeLa, HEK293, and SKNMC cells. Confocal microscopy analysis shows that pepMAX can efficiently deliver mRNA into the cytosol and induce efficient protein production. Furthermore, the pepMAX/mRNA co-assemblies retain their transfection efficiency even after being stored in lyophilized form for one week at room temperature.
