Imaging Flow Cytometry Demonstrates Physiological and Morphological Diversity within Treated Probiotic Bacteria Groups

Summary: Probiotic bacteria can be introduced to stresses during culturing phase as a substitute for protectants and coating substances during drying. Accurate bacterial count enumeration in probiotic formulation can be achieved using imaging flow cytometry (IFC), which combines statistical power of conventional flow cytometry with imaging content of microscopy. IFC provides more information by correlating fluorescence light data with digitally processed images, offering additional insight into cell physiology and morphology compared to standard methods (plate cell counts and traditional flow cytometry). IFC with complementary dyes allows for grouping based on metabolic activity and membrane damage, and cell sorting is used to assess growth on different media. Results show viable but nonculturable cells correspond to groups with intermediate metabolic activity and some degree of cellular damage.

Summary: This study investigated the probiotic, hydrophobic, and immune modulation properties of Streptococcus lactarius MB622 and Streptococcus salivarius MB620 isolated from human milk. Results showed that both strains displayed high hydrophobicity and had probiotic properties such as gram-positive classification, catalase negative activity, resistance to gastric juice, and bile salt concentration. Administering these strains in sufficient amount and for a certain duration could reduce inflammation in the colon by decreasing the production of inflammatory booster (IL-8) in diseased state.
