Maternal obesity and programming of metabolic syndrome in the offspring: searching for mechanisms in the adipocyte progenitor pool

Taylor B. Scheidl et al.

Summary: Recent studies have shown that the quality of adipose tissue, rather than its quantity, contributes to the risk of obesity-related diseases. Adipose-derived stem cells give rise to adipocytes during the development of adipose depots. In adult depots, progenitor cells serve as a reservoir for replacing aging adipocytes and increasing lipid storage capacity.

Prader-Willi Syndrome and Chromosome 15q11.2 BP1-BP2 Region: A Review

Merlin G. Butler

Summary: Prader-Willi syndrome (PWS) is a complex genetic disorder characterized by hypotonia, hypogonadism, and developmental delay in infancy. In childhood, it presents with obesity, learning and behavioral problems, and hormone deficiencies. Severe symptoms are associated with the deletion of four non-imprinted genes (NIPA1, NIPA2, CYFIP1, TUBGCP5) from the 15q11.2 BP1-BP2 region. These genes are involved in brain and muscle development, glucose and insulin metabolism, and neurobehavioral outcomes. Deletion of the 15q11.2 BP1-BP2 region alone can lead to Burnside-Butler syndrome, which includes neurodevelopmental, motor, learning, and behavioral problems.

Stromal Vascular Fraction Obtained From Subcutaneous Adipose Tissue: Ex-Obese and Older Population as Main Clinical Targets

Francisca Frias et al.

Summary: This study investigates the correlation between age, body mass index (BMI), history of obesity and massive weight loss, and harvest site with the expression of stromal vascular fraction (SVF) cell markers in human adipose tissue. The results indicate that ex-obese patients have a higher percentage of SVF cells expressing CD90. BMI, harvest site, and age do not appear to be associated with SVF subpopulations.

Adipose-tissue plasticity in health and disease

Alexander Sakers et al.

Summary: Adipose tissue, known as fat, plays a crucial role in regulating various physiological aspects of the body. Its high plasticity allows for significant changes in metabolism, structure, and phenotype in response to physiological stimuli. Limitations in this plasticity contribute to decreased or aberrant responses to physiological cues, leading to the progression of cardiometabolic disease.

Diverse effect of MC4R risk alleles on obesity-related traits over a lifetime: Evidence from a well-designed cohort study

Maryam Moazzam-Jazi et al.

Summary: This study is the first to investigate the association of common MC4R SNPs with obesity-related parameters, revealing several SNPs that are associated with obesity indicators in Iranian adults. An interesting finding is a protective gene that helps combat obesity. Additionally, the MC4R risk score only affects BMI and waist circumference during early adulthood in the study.

Adipose tissue aging: mechanisms and therapeutic implications

Min-Yi Ou et al.

Summary: Adipose tissue undergoes significant changes during aging, leading to physiological declines and age-related diseases in the elderly population. These changes involve redistribution of fat deposits, decline in the function of adipocyte progenitors, and accumulation of senescent cells. Pathogenic pathways induce adipose tissue dysfunction, which can result in chronic inflammation, metabolic system disruption, and acceleration of the aging process.

The results showed that microfragmented adipose tissue had a higher population of endothelial progenitors (EP) compared to leukocytes and supra-adventitial-adipose stromal cells (SA-ASC). The ratio of progenitor cells differed between male and female patients, with men showing a higher EP-pericyte and pericyte-SA-ASC ratio.

Summary: In Prader-Willi syndrome (PWS) patients, abnormal white adipose tissue (WAT) expansion and hypertrophy occur before the onset of obesity, accompanied by increased apoptosis. The adipogenic programs for both white and beige adipocytes are impaired in PWS patients, as evidenced by RNA-seq analysis. SNORD116 deficiency contributes to disrupted adipogenesis. These findings expand our understanding of the pathogenesis of PWS obesity and may contribute to the development of new therapeutic strategies.

BBS4 Is Essential for Nuclear Transport of Transcription Factors Mediating Neuronal ER Stress Response

Avital Horwitz et al.

Summary: Bardet-Biedl syndrome (BBS) is an autosomal recessive syndrome characterized by retinal dystrophy, cognitive impairment, and obesity. Our study focused on examining the role of BBS4 in neuronal development under normal and ER stress conditions. BBS4 depletion in neuronal cells leads to reduced sensitivity to ER stress during differentiation, partly due to the failure in translocation of ER-transcription factors (TF) sXBP-1 and ATF6 alpha p50 to the nucleus.
