Mechanistic Understanding of D-Glucaric Acid to Support Liver Detoxification Essential to Muscle Health Using a Computational Systems Biology Approach

V. A. Shiva Ayyadurai et al.

Summary: Liver and muscle health are closely related, and nutritional strategies that support liver detoxification can benefit muscle recovery. A computational analysis of calcium and potassium glucarate salts and their metabolite D-glucaric acid reveals their positive effects on liver detoxification through multiple molecular pathways. By downregulating apoptosis, reducing glucuronide deconjugates levels, reducing ROS production, and inhibiting beta-glucuronidase enzyme, D-glucaric acid improves liver detoxification. This in silico study provides a mechanistic explanation for the mitigation of liver toxicity by D-glucaric acid.

Paradigms of endothelial stiffening in cardiovascular disease and vascular aging

Victor M. M. Aguilar et al.

Summary: Endothelial cells, the inner lining of blood vessels, have a critical role in vascular function, but endothelial dysfunction contributes to cardiovascular disease. This review focuses on endothelial stiffness, which reflects the cell's resistance to deformation. The mechanisms and factors that determine and induce endothelial stiffening are discussed, including RhoA-dependent contraction, actin architecture, mechanical signals, and lipid factors. The contribution of endothelial stiffening in cardiovascular disease and aging, as well as its impact on endothelial barrier disruption, are also examined.

Aging Hallmarks and the Role of Oxidative Stress

Edio Maldonado et al.

Summary: Aging is a complex biological process with a decline in physical function and increased risk of chronic diseases. Studies have identified nine hallmarks of aging, including telomere shortening, genomic instability, epigenetic modifications, mitochondrial dysfunction, loss of proteostasis, dysregulated nutrient sensing, stem cell exhaustion, cellular senescence, and altered cellular communication. Among these mechanisms, mitochondrial dysfunction and the resulting elevated reactive oxygen species (ROS) production play a crucial role in age-related damage and the aging phenotype.

Food Anthocyanins: Malvidin and Its Glycosides as Promising Antioxidant and Anti-Inflammatory Agents with Potential Health Benefits

Anna Merecz-Sadowska et al.

Summary: Anthocyanins are flavonoid compounds found in fruits and vegetables, which contribute to their color and offer various health benefits due to their biological properties. Malvidin, one of the most well-known anthocyanidins, and its glycosides have been shown in multiple studies to possess anti-carcinogenic, diabetes-control, cardiovascular-disease-prevention, and brain-function-improvement properties. These health benefits are primarily attributed to their antioxidant and anti-inflammatory effects, which are influenced by the modulation of critical genes. This article reviews the biological activity of malvidin and its glycosides and their role in promoting health.
