Reductive Ring Opening of Arylcyclopropanecarboxamides Accompanied by Borylation and Enolate Formation

Shuo Wang et al.

Summary: In this study, treatment of arylcyclopropanecarboxamides with a sodium dispersion in the presence of methoxypinacolborane as a reduction-resistant electrophile was investigated. The results showed that the reaction led to the reductive cleavage of the cyclopropane ring followed by instant trapping with the boron electrophile, resulting in the formation of enolates of gamma-aryl-gamma-borylalkanamides. The enolates could further react with a different electrophile to produce the corresponding a-substituted amides with anti selectivity.

Synthesis of Imidized Cyclobutene Derivatives by Strain Release of [1.1.1]Propellane

Xiaofan Du et al.

Summary: In this study, a metal-free synthesis of imidized methylene cyclobutane derivatives was reported using a strain-release driven addition reaction of [1.1.1]propellane. The methylene cyclobutyl cation intermediate generated by protonation of [1.1.1]propellane was trapped by nitriles to form a nitrilium ion intermediate, which subsequently reacted with carboxylic acids to produce imidized methylene cyclobutene derivatives via a Mumm-type rearrangement.

Reactivity of 1.1.1-Propellane with (silox)3M (M = Ti, V, Cr): Structures of (silox)3V=(cC4H4)=CH2 and [(silox)3Cr-(1.1.1-C5H6)-]2

Gregory M. George et al.

Summary: The two-orbital, two-electron, four-state paradigm was applied to 1.1.1-propellane with an unusual "inverted" central bond, indicating that an electrophilic metal center can activate it towards ring opening. Low-valent, early-metal complexes resulted in different reactions, with facile ring opening in some cases and radical-like dimerization in others. High-level calculations provided a rationale for the trichotomy in reactivity observed in the experiments.

Visible-light-mediated multi-component carbene transfer reactions of α-diazoesters to construct multisubstituted pyrazoles and 1,3-dicarbonyl derivatives

Hongyu Ding et al.

Summary: A visible-light-mediated strategy for the synthesis of multisubstituted pyrazoles through a three-component carbene transfer reaction of alpha-diazoesters with cyclic ethers and pyrazolones is reported. This transformation can be achieved under mild and additive-free conditions, yielding a wide range of multisubstituted pyrazoles with moderate to good yields and excellent functional group compatibility. Additionally, a series of substituted 1,3-dicarbonyl derivatives can be conveniently obtained via visible-light-promoted three-component reaction of alpha-diazoesters, cyclic ethers, and 1,3-dicarbonyl compounds.

Electrophilic Activation of [1.1.1]Propellane for the Synthesis of Nitrogen-Substituted Bicyclo[1.1.1]pentanes

Sarah Livesley et al.

Summary: The study presents a method for electrophilic activation of [1.1.1]propellane in a halogen bond complex, allowing its reaction with electron-neutral nucleophiles to yield nitrogen-substituted BCPs. Computational analysis reveals that the halogen bonding interaction promotes nucleophilic attack without sacrificing cage stabilization. Overall, the work demonstrates the rehabilitation of electrophilic activation of [1.1.1]propellane as a valuable strategy for accessing functionalised BCPs.

Visible-Light-Induced 1,3-Aminopyridylation of [1.1.1]Propellane with N-Aminopyridinium Salts

Summary: The formation of an electron donor-acceptor complex allowed for the direct installation of amino and pyridyl groups onto bicyclo[1.1.1]pentane (BCP) frameworks in the absence of an external photocatalyst. This method is robust and metal-free, enabling late-stage modification of structurally complex biorelevant molecules. The strategy also facilitates the incorporation of functional groups with pyridine across the BCP core, significantly expanding the range of potential applications for drug discovery.

Recent Advances in Theoretical Studies on Transition-Metal-Catalyzed Carbene Transformations

Xiaotian Qi et al.

Summary: Metal carbenes play a crucial role in modern organic synthesis, and computational studies of their transformation reactions with different nucleophiles can provide valuable mechanistic insights for the design of new reactions. Three generalized reaction models for carbene transformation with nucleophiles are proposed based on our computational results: intramolecular migratory insertion of metal carbene, intermolecular nucleophilic addition towards metal carbene, and outer-sphere nucleophilic addition to metal-ligated free carbene.

Bridge Functionalisation of Bicyclo[1.1.1]pentane Derivatives

Joseph M. Anderson et al.

Summary: By replacing aromatic rings, internal alkynes, and tert-butyl groups with bicyclo[1.1.1]pentane (BCP) units, the saturation level and three-dimensionality of drug-like compounds can be increased to enhance their potency and selectivity. While functionalization of the tertiary bridgehead positions of BCP derivatives is well-studied, functionalization of the three concyclic secondary bridge positions is still a developing field. The unique properties of the BCP core present synthetic challenges but offer novel vectors for drug discovery and applications in materials science.
