Comprehensive Mechanistic Scenario for the Cu-Mediated Asymmetric Propargylic Sulfonylation Forging Tertiary Carbon Stereocenters

Summary: Metal-catalyzed propargylic transformations are a powerful tool in organic synthesis for forming new carbon-carbon and carbon-heteroatom bonds. However, there is limited knowledge about the mechanistic intricacies of asymmetric propargylic reactions with challenging heteroatom substitution, presenting an inspiring challenge. This study presents a detailed mechanistic analysis of a propargylic sulfonylation reaction promoted by a chiral Cu catalyst through experimental techniques and computational studies. Surprisingly, the enantio-discriminating step was found to be the proto-demetalation step, rather than the coupling between the nucleophile and the propargylic precursor.

Copper-Catalyzed Diastereo- and Enantioselective Decarboxylative [3+2] Cyclization of Alkyne-Substituted Cyclic Carbamates with Azlactones: Access to γ-Butyrolactams Bearing Two Vicinal Tetrasubstituted Carbon Stereocenters

Summary: A copper-catalyzed decar-boxylative [3 + 2] cyclization reaction between alkyne-substituted cyclic carbamates and azlactones has been developed, providing optically pure gamma-butyrolactams bearing two adjacent tetrasubstituted carbon stereocenters in high yields with excellent stereo-selectivities (up to 99% yield, 99:1 dr, and 99% ee). This represents the first successful asymmetric synthesis of gamma-butyrolactams containing sterically congested adjacent tetrasubstituted stereocenters via a decarboxylative cyclization pathway.

Summary: This study demonstrates the efficient chiral aldehyde-nickel dual catalytic system for the direct asymmetric alpha-propargylation reaction of amino acid esters with propargylic alcohol derivatives, leading to the synthesis of structurally diverse α,α-disubstituted non-proteinogenic α-amino acid esters. The stereodivergent synthesis of natural product NP25302 is also achieved, and a reasonable reaction mechanism is proposed based on control experiments, nonlinear effect investigation, and HRMS detection.

Ruthenium-Catalyzed Enantioselective Propargylic Phosphinylation of Propargylic Alcohols with Phosphine Oxides

Summary: In recent years, significant progress has been made in the development of transition metal-catalyzed enantioselective propargylic substitution reactions. However, until now, no successful example with phosphorus-centered nucleophiles has been reported. This study presents the first successful example of ruthenium-catalyzed enantioselective propargylic substitution reactions using propargylic alcohols as substrates and diarylphosphine oxides as phosphorus-centered nucleophiles, providing a new method for preparing chiral phosphorus-containing organic compounds.
