Genome mining for unknown-unknown natural products

Danielle A. A. Yee et al.

Summary: Genome mining revealed a conserved fungal biosynthetic pathway that lacked a core enzyme. Heterologous expression identified a highly modified cyclo-arginine-tyrosine dipeptide as the associated natural product. Biochemical characterization led to the discovery of a new arginine-containing cyclodipeptide synthase (RCDPS) that showed no sequence homology to known enzymes and is widely encoded in fungal genomes. Further research on the biosynthetic pathway led to the discovery of new compounds whose structures would not have been predicted without the knowledge of RCDPS function.

Canavanine utilization via homoserine and hydroxyguanidine by a PLP-dependent γ-lyase in Pseudomonadaceae and Rhizobiales

Franziskus Hauth et al.

Summary: A novel PLP-dependent enzyme, canavanine-gamma-lyase, has been characterized to catalyze the hydrolysis of canavanine to homoserine. This enzyme induction also facilitates canavanine utilization by inducing the activities of homoserine-dehydrogenase, aspartate-semialdehyde-dehydrogenase, and ammonium-aspartate-lyase. The findings expand the range of reactions that PLP-dependent enzymes can catalyze and could have important implications for the establishment and maintenance of the legume rhizosphere.

L-Threonine Transaldolase Activity Is Enabled by a Persistent Catalytic Intermediate

Prasanth Kumar et al.

Summary: L-Threonine transaldolases (LTTAs) are a class of enzymes dependent on PLP, responsible for synthesizing diverse beta-hydroxy amino acids. ObiH, an essential LTTA for the biosynthesis of the beta-lactone natural product obafluorin, has been studied to understand its catalytic mechanism and structural features. The data explain the unique reactivity of LTTA enzymes and provide a basis for future engineering and mechanistic analysis.

A shared mechanistic pathway for pyridoxal phosphate-dependent arginine oxidases

Elesha R. Hoffarth et al.

Summary: Recent study uncovers a mechanism by which the organic cofactor PLP activates molecular oxygen for oxidation reactions, particularly in the desaturation or hydroxylation of arginine. The proposed mechanism suggests the precise positioning of a water molecule in the active site plays a crucial role in controlling the final reaction outcome. This mechanism provides a unified framework to understand the activation of oxygen by PLP-dependent enzymes and elucidates a shared mechanistic pathway and intertwined evolutionary history for arginine desaturases and hydroxylases.
