Genome-wide investigation of multiplexed CRISPR-Cas12a-mediated editing in rice

Summary: Clustered regularly interspaced short palindromic repeats (CRISPR) nucleases, such as Cas9 and Cas12a, are revolutionizing plant research and breeding by enabling multiplexed genome editing. In this study, the potential off-target effects of inducing multiple DNA double-strand breaks (DSBs) in a crop genome using a highly multiplexed CRISPR-Cas12a system were investigated in rice. The results revealed large chromosomal rearrangement events in rice plants enduring many simultaneous DSBs, providing important insights into the analysis and regulation of engineered crops derived from CRISPR-Cas mediated multiplexed genome editing.

This article reviews the historical aspects of plant metabolic gene cluster studies and provides general strategies for identifying plant metabolic gene clusters in the era of multi-omics. The exploration of natural variation and evolution, as well as the understanding of how the structure of plant metabolic gene clusters influences their function, are also emphasized.

Cold acclimation diversity in Arabidopsis thaliana: CRISPR/Cas9 as a tool to fine analysis of Tandem Gene Arrays, application to CBF genes
