FIBER-ML, an Open-Source Supervised Machine Learning Tool for Quantification of Fibrosis in Tissue Sections

Summary: Pathologic fibrosis is a major characteristic of tissue insult in chronic diseases, but there is currently no consistent method for its quantification. This study tested a new open-source software, FIBER-ML, which can automatically quantify fibrosis after a learning phase. The software performed well and showed excellent correlation and agreement with other software. It provides a user-friendly alternative with quality control and file management.

Cardiomyocyte protein O-GlcNAcylation is regulated by GFAT1 not GFAT2

Summary: In response to cardiac injury, the hexosamine biosynthesis pathway (HBP) plays a significant role, with glutamine-fructose amidotransferase (GFAT) being a key enzyme in the pathway. GFAT1 is primarily expressed in cardiac myocytes, while GFAT2 is mainly expressed in cardiac fibroblasts. Knockdown of GFPT1 in cardiac cells resulted in reduced HBP activity. Cell-specific isoform expression may have different effects on cell function, which should be considered in studies of HBP and GFAT functions in the heart.

Segmental analysis by speckle-tracking echocardiography of the left ventricle response to isoproterenol in male and female mice

Summary: The study found that isoproterenol negatively affected left ventricular function in male and female mice, reducing ejection fraction and global and segmental strain. Male mice showed more severe modulation in the LV posterior segments, while both anterior and posterior wall segments were impacted in females. Although diastolic function parameters returned to normal after cessation of isoproterenol, LV dilation and decreased ejection fraction persisted in males even 17 weeks post-treatment.

The Hexosamine biosynthesis pathway (HBP) plays a key role in mediating cardiac hypertrophy through protein O-GlcNAcylation and Akt activation. GFAT2, a critical enzyme in the HBP pathway, is identified as a major isoform in the heart and is involved in regulating cardiomyocyte hypertrophy.

Sacubitril/valsartan decreases mortality in the rat model of the isoprenaline-induced takotsubo-like syndrome

Summary: This study aimed to investigate the effect of sacubitril/valsartan in preventing isoprenaline-induced TTS-like phenotype in rats. The results showed that pretreatment with sacubitril/valsartan reduced mortality and attenuated cardiac dysfunction in the TTS-like model induced by isoprenaline. This suggests that sacubitril/valsartan could be a potential treatment option for patients with TTS.
