Tau interactome maps synaptic and mitochondrial processes associated with neurodegeneration

Early prediction of preeclampsia in pregnancy with cell-free RNA

Summary: Liquid biopsies that measure circulating cell-free RNA can be used to study the development of pregnancy-related complications. This study identified and validated cfRNA transcriptomic changes associated with preeclampsia, and found that these changes are marked and stable early in gestation. Genes specific to neuromuscular, endothelial, and immune cell types and tissues are enriched in these changes, suggesting their relevance to preeclampsia physiology and providing new hypotheses for disease progression. Based on these findings, a panel of 18 genes was identified that can form the basis of a liquid biopsy test to identify mothers at risk of preeclampsia long before symptoms occur.

Summary: Autophagy is a crucial process in maintaining normal pregnancy, but it is impaired in preeclampsia. This study demonstrates that hypoxia-reoxygenation treatment disrupts the autophagic machinery in human trophoblasts, leading to the accumulation of protein aggregates. Similar features were observed in the placenta from early-onset preeclampsia deliveries. Evaluation of key regulators and verification of autophagic ultrastructure are essential for accurately assessing autophagy activity in human trophoblasts and placental tissue from preeclampsia deliveries.

Cerebral Biomarkers and Blood-Brain Barrier Integrity in Preeclampsia

Summary: Cerebral complications in preeclampsia are a significant threat to maternal health and survival. Currently, there is a lack of reliable and accessible predictors for preeclampsia-related cerebral complications. This study found that plasma concentrations of the cerebral biomarkers NfL, tau, NSE, and S100B were all higher in women with preeclampsia compared to women with normal pregnancies and non-pregnant women. In addition, higher plasma concentrations of NfL were associated with decreased blood-brain barrier integrity in an in vitro model.
