INCA 2.0: A tool for integrated, dynamic modeling of NMR- and MS-based isotopomer measurements and rigorous metabolic flux analysis

Mohsin Rahim et al.

Summary: Metabolic flux analysis is a method that combines experimental measurements and computational modeling to determine reaction rates in biological systems. This study introduces a software tool called INCA 2.0, which can integrate isotopomer measurements from both MS and NMR platforms and estimate metabolic fluxes with flexibility. The results demonstrate that INCA 2.0 accurately simulates and analyzes NMR datasets, outperforming other established tools. Additionally, combining NMR and MS datasets improves the precision of estimated fluxes.

Increased vascular permeability and severe renal tubular damage after ischemia-reperfusion injury in mice lacking adiponectin or T-cadherin

Yuri Tsugawa-Shimizu et al.

Summary: The study showed that APN protects the kidney in an acute renal injury model by binding to T-cadherin. T-cadherin-associated adiponectin protein exists on peritubular capillary pericytes, and deficiency of both leads to more severe renal tubular damage and increased vascular permeability. These findings suggest that APN and T-cadherin play an important role in maintaining the capillary network and ameliorating renal tubular injury.

High-Resolution Human Kidney Molecular Histology by Imaging Mass Spectrometry of Lipids

Summary: The study demonstrates the potential of imaging mass spectrometry in lipidomics to highlight the structural features of the human kidney at a spatial resolution of 10 μm. Specific lipid fingerprints help identify different segments of the nephron and interstitial tissue, laying the foundation for further research on the metabolic basis of diseases affecting the human kidney.

Summary: The discovery of FAM195A, a BAT-enriched RNA binding protein, highlights its essential role in cold-dependent thermogenesis in mice. Knockout of FAM195A results in whitening of BAT and impaired thermoregulation, affecting the response to cold significantly.

Multitissue 2H/13C flux analysis reveals reciprocal upregulation of renal gluconeogenesis in hepatic PEPCK-C-knockout mice

Mohsin Rahim et al.

Summary: The study demonstrates that the kidney can compensate for impaired hepatic glucose production by increasing renal glucose production and gluconeogenesis. Hepatic gluconeogenesis was reduced in KO mice, but whole-body glucose production and arterial glucose were unaffected, leading to a compensatory rise in renal glucose production and gluconeogenesis.

Quantitative flux analysis in mammals

Caroline R. Bartman et al.

Summary: The article provides a detailed review of techniques for measuring metabolic fluxes in intact mammals and emphasizes the importance of measuring metabolism in vivo. Researchers studying glucose homeostasis have developed strategies that have been strengthened by recent advances in metabolomics technologies, with promising avenues for future application. The broader application of these methods has the potential to accelerate biomedical progress given the significant importance of metabolism to health and disease.
