circPTEN1, a circular RNA generated from PTEN, suppresses cancer progression through inhibition of TGF-β/Smad signaling

Lin Zheng et al.

Summary: This study revealed a novel circular RNA (circPTEN1) generated from the PTEN gene, which is downregulated in colorectal cancer and associated with poor survival. Low expression of circPTEN1 promotes metastasis and invasion, while overexpression shows opposite effects. Mechanistically, circPTEN1 binds to Smad4 and disrupts its interaction with Smad2/3, inhibiting the formation and translocation of Smad complexes. Additionally, eIF4A3 suppresses the cyclization of circPTEN1.

Summary: Esophageal squamous cell carcinoma (ESCC) is a highly aggressive gastrointestinal cancer. Identifying sensitive and specific biomarkers for ESCC detection and treatment is crucial. Circular RNAs (circRNAs) have a unique circular structure, making them more stable and potentially valuable prognostic and diagnostic biomarkers for ESCC. Multiple studies have shown the important roles of circRNAs in regulating ESCC cells. This article summarizes the current progress in aberrantly expressed circRNAs, their biological functions, clinical significance in ESCC, and introduces databases for circRNA research. With advancements in high-throughput sequencing and bioinformatics techniques, circRNAs offer new possibilities for precision treatment of ESCC.

Breast Cancer Statistics, 2022

Angela N. Giaquinto et al.

Summary: This article provides an update on female breast cancer statistics in the United States, including data on incidence, mortality, survival, and mammography screening. It shows that breast cancer incidence rates have increased over the past few decades, driven by localized-stage and hormone receptor-positive disease. However, breast cancer mortality rates have been steadily declining since 1989, although at a slower pace in recent years. Despite lower incidence rates, there remains a racial disparity in breast cancer mortality, with higher rates among Black women compared to White women.
