UHRF1 inhibition epigenetically reprograms cancer stem cells to suppress the tumorigenic phenotype of hepatocellular carcinoma

Lenvatinib inhibited HCC cell migration and invasion through regulating the transcription and ubiquitination of UHRF1 and DNMT1

Summary: In this study, it was found that Lenvatinib can inhibit the metastasis and epithelial-mesenchymal transition of hepatocellular carcinoma cells. This effect is achieved by negatively regulating the transcription of UHRF1 and DNMT1 through the ERK/MAPK pathway, as well as promoting their protein degradation through the ubiquitin-proteasome pathway. Consequently, the expression of E-Cadherin is upregulated and the adhesion and metastasis of Huh7 cells are attenuated in vivo.

Recent studies have found differences in cancer stem cell properties among different liver cancer types. Liver cancer stem cells, also known as liver tumor-initiating cells, are considered to be drivers of tumor initiation and metastasis, and are regulated by various mechanisms and factors. This review presents an analysis of cancer stemness in different liver cancer types, evaluates the mechanisms of cancer stemness maintenance in liver cancer stem cells, and discusses potential treatments for eradicating liver cancer stem cells.

Targeting UHRF1-SAP30-MXD4 axis for leukemia initiating cell eradication in myeloid leukemia

Summary: Aberrant expression of UHRF1, an epigenetic regulator, in acute myeloid leukemia (AML) is associated with poor prognosis. UHRF1 is crucial for the self-renewal of leukemia initiation cells (LICs) and its interaction with SAP30 represses gene expression. Inhibition of UHRF1 or SAP30 leads to derepression of MXD4, an MYC antagonist, and suppresses leukemogenesis. Furthermore, a UHRF1 inhibitor, UF146, shows promising therapeutic efficacy in a myeloid leukemia model.

Inhibition of PGC7/GLI1/MYCN might reverse poorly differentiated HCCs and provide new therapeutic strategies.

These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

uhrf1 and dnmt1 Loss Induces an Immune Response in Zebrafish Livers Due to Viral Mimicry by Transposable Elements

Summary: The activation of transposable elements (TEs) can lead to cellular damage, where cytoplasmic nucleic acid sensing pathways play a role in not only detecting pathogens but also eliminating cells with inappropriate TE activation. DNA methylation plays a direct role in suppressing certain subclasses of TEs, while the induction of interferon response and tnfa activation in liver cells of mutant organisms mimics gene expression patterns seen in virally infected cells. Antiviral response induced by DNA hypomethylation and TE activation in the liver is mediated by signaling pathways activated by both cytoplasmic double stranded RNA and DNA, with tnfa potentially mediating cell death as a mechanism to eliminate damaged cells.
