Summary: The ongoing global Monkeypox outbreak highlights the importance of live virus vaccines based on vaccinia virus (VACV) to protect the population. There is a need for a safer and more effective vaccine against both Monkeypox and Smallpox. One potential approach is the use of horsepox virus (HPXV) as a vaccine, which shares a common ancestor with VACV. A proof-of-concept study shows that TNX-801, a recombinant chimeric horsepox virus, elicited protective immune responses against Monkeypox virus.

Molecular Mechanisms of Poxvirus Evolution

Summary: Poxviruses, despite being double-stranded DNA pathogens with proofreading polymerases, have highly adaptable genomes that can rapidly undergo genotypic and phenotypic changes. Recent increase in human-to-human transmission of monkeypox virus serves as an example of this adaptability. Advances in deep sequencing technologies have revealed standing nucleotide variation in poxvirus populations that was previously underestimated. There is also growing understanding of the role of genomic architectural changes in shaping poxvirus evolution, including recombination, gene duplications and loss, and acquisition of new genes through horizontal gene transfer.

Cowpox Viruses: A Zoo Full of Viral Diversity and Lurking Threats

Summary: In this article, we review the history of the evolution of cowpox viruses (CPXVs) and their broad host range, lethal outbreaks, and increasing human cases in Eurasian countries. Genetic analyses have revealed multiple Orthopoxvirus species categorized under CPXV designation. We discuss modern outbreaks in zoos, domesticated animals, and humans, including the pathogenesis, geographic range, and molecular level interactions. The potential threat of these viruses and the future of CPXV research are also discussed to provide a comprehensive review of CPXVs.

Genomic Sequencing and Analysis of a Novel Human Cowpox Virus With Mosaic Sequences From North America and Old World Orthopoxvirus

Summary: In this study, the complete genome sequence of an atypical cowpox virus (CPXV-No-H2) isolated from a human in Norway was determined. The virus showed a mosaic genome with genes most similar to other orthopoxviruses and multiple potential recombination events were identified. Phylogenetic analysis revealed that CPXV-No-H2 formed a distinct clade with German CPXV isolates and clustered closely with the ECTV-OPXV Abatino clade.

Poxviruses, like Orthopoxviruses (ORPV), have a significant number of genes dedicated to counteracting host immunity. Through evolution, ORPV acquired accessory genes in three major waves, leading to gene duplication and the formation of gene families. Interestingly, most accessory genes were lost during ORPV evolution, suggesting a variety of functional interactions within this virus family.
