Novel plant-derived exosome-like nanovesicles from Catharanthus roseus: preparation, characterization, and immunostimulatory effect via TNF-α/NF-κB/PU.1 axis

Summary: This study isolated novel CLDENs from Catharanthus roseus leaves that showed excellent stability and biocompatibility and can be used for post-chemotherapy immune adjuvant therapy. Lipidomic and differential proteomic analysis revealed the unique lipid composition and origin of CLDENs, and six marker proteins were identified. In vitro and in vivo experiments demonstrated that CLDENs promoted macrophage polarization and phagocytosis, lymphocyte proliferation, and had anti-inflammatory and hematopoietic functions. Plant cell culture system was established to obtain gram-level nanovesicles with similar properties and activities as CLDENs.

Milk-derived extracellular vesicles protect intestinal barrier integrity in the gut-liver axis

Summary: Milk-derived extracellular vesicles (mEVs) show similar protective effects on intestinal barrier integrity as well as the prevention of endotoxin translocation into the liver in gut inflammation and diet-induced metabolic diseases. Oral administration of mEVs restores gut barrier integrity and alleviates gut disorders and their associated liver inflammation, suggesting that mEVs have potential in the treatment of gut-liver axis-associated metabolic diseases via protection of intestinal barrier integrity.
