Global prevalence, incidence, and outcomes of alcohol related liver diseases: a systematic review and meta-analysis

Xuanxuan Niu et al.

Summary: This review investigated the global prevalence of alcohol-related liver disease (ARLD) and found that it is 4.8%. The prevalence is higher in males and Caucasians. Alcoholic liver cirrhosis is the most common type of ARLD, and ascites is the most common complication. The incidence rate of ARLD is 0.208/1000 person-years, and the overall mortality rate is 23.9%, with liver-related mortality at 21.6%. Urgent measures are needed to reduce the factors contributing to the high disease prevalence.

Recent Advances in Understanding of Pathogenesis of Alcohol-Associated Liver Disease

Xiaoqin Wu et al.

Summary: Alcohol-associated liver disease (ALD) is a significant disease caused by chronic alcohol consumption, which is also one of the most common causes of liver-related morbidity and mortality. It includes various stages such as liver steatosis, fibrosis, cirrhosis, and alcohol-associated hepatitis. The development of ALD involves complex interactions between cells and organs, affecting homeostatic mechanisms, injury and repair regulation in the liver. Recent advances have been made in understanding the disruption of programmed cell death and prosurvival pathways, innate immune responses, liver sinusoidal endothelial cell dysfunction, and gut-liver and adipose-liver cross talk in response to ethanol. Potential therapeutic targets and clinical trials for the treatment of ALD, including alleviating hepatocyte injury, reducing inflammatory responses, and targeting gut microbiota, are also discussed.

The Global Burden of Liver Disease

Zobair M. Younossi et al.

Summary: Chronic liver disease and its associated complications cause significant mortality, morbidity, and economic burden. The burden of CLD is large and increasing globally, with nonalcoholic fatty liver disease and alcohol-related liver disease being major contributors. Different regions of the world are affected by different etiologies, with HBV-related HCC cases being highest in Asia and NAFLD-related HCC cases highest in the United States. The impact of CLD is influenced by sociodemographic factors.

Flavonoids and cellular stress: a complex interplay affecting human health

Hanne Vissenaekens et al.

Summary: Flavonoid consumption is beneficial to human health, but clinical evidence is often inconclusive due to high interindividual variability. Research on the bidirectional interplay between flavonoids and cellular stress, as well as the influence of cellular stress on flavonoid uptake and responses, is crucial for establishing personalized food recommendations. Further development of cell models that simulate physiological micro-environments with interindividual variability is also needed for future mechanistic research.

Anti-Inflammatory Activity of Mulberry Leaf Flavonoids In Vitro and In Vivo

Ziwei Lin et al.

Summary: This study found that mulberry leaf flavonoids (MLFs) eluted by 30% ethanol exhibited the best anti-inflammatory and antioxidative activities. In cell experiments, MLFs effectively inhibited the production of inflammatory cytokines. Moreover, these MLFs also enhanced antioxidative capacity. Untargeted metabolomic analysis revealed that quercetin and kaempferol compounds were more abundant in the 30% MLFs. Animal experiments confirmed that 30% MLF can alleviate symptoms of ulcerative colitis and inhibit the activation of the inflammatory pathway.
