IGFBP3 epigenetic promotion induced by METTL3 boosts cardiac fibroblast activation and fibrosis

Ji-Fei Ding et al.

Summary: Cardiac fibrosis, caused by activation and proliferation of cardiac fibroblasts, remains a challenging problem in heart disease. The gene IGFBP3 was found to be significantly upregulated in cardiac fibrosis, and its knockdown inhibited migration and proliferation of fibroblasts. Additionally, abnormal N6-methyl-adenosine(m6A) mRNA modifications were found to regulate IGFBP3 expression in developing cardiac fibrosis. METTL3 may regulate IGFBP3 expression and fibroblast activation through RNA epigenetic modifications, providing a potential therapeutic target.

Summary: Through bioinformatics analysis, this study identified 130 differentially expressed immune-related genes (DEIRGs) that play crucial roles in immune and inflammatory responses. Immune infiltration analysis revealed that LA tissues from AF-VHD patients have higher levels of gamma delta T cells but lower levels of CD8 and regulatory T cells compared to SR patients. Key DEIRGs showed correlations with various infiltrating immune cells, indicating their involvement in the immune response in AF-VHD pathogenesis.
