Solid-state fermentation conditions optimization, homology modelling and molecular docking of β-mannanase of a novel Streptomyces species LB66 isolated from Sargassum seaweed waste

Summary: This study optimized the solid-state fermentation conditions using cow dung as the feedstock for maximal production of beta-mannanase. The 3D structure and enzyme-substrate interactions of the modelled beta-mannanase were elucidated using homology modelling and molecular docking methods. The identified active site residues can be targeted for directed evolution approaches to tailor enzymatic traits and enable ecofriendly bioprocessing.

Summary: This study investigated the physicochemical properties of purified endo-1,4-beta-mannanase from the wild type, Alcaligenes sp. and its most promising chemical mutant. The wild and mutant enzymes were purified and analyzed for their molecular weights, optimal temperature and pH, stability, and substrate affinity. The results showed that the mutant enzyme had improved thermo-stability, thermo-tolerant, and high substrate affinity compared to the wild-type enzyme. These findings have important implications for enhancing bioprocesses.

The enzyme belongs to GH family 5 and subfamily 7, with a molecular weight of 40.6 kDa. It has optimum temperature and pH at 70°C and 5.5, respectively, and shows high stability at 50°C. The enzyme is sensitive to Mn+2, Hg+2, and Co+2, but stimulated by Zn+2.

Effects of dietary ß-mannanase (Hemicell®) and Lavandula angustifolia on Oreochromis niloticus fed a low level of dietary protein: Growth, digestive enzymes, and hemato-biochemical indices

Summary: High feed prices and limited access to feed are significant challenges for fish producers. The study explored the potential use of Hemicell (R) (HC) and lavender flower meal (LFM) as additives in a low-protein diet for Oreochromis niloticus. The results showed that the addition of HC alone or with LFM improved growth, feed utilization, and digestive enzyme activity compared to the control group. However, no major impacts were observed on water quality or fish health indicators. This study suggests that adding HC to low-protein diets could be a cost-effective solution for the aquaculture industry.
