Summary: This study developed and validated a multiplex immunoassay (MIA) for the simultaneous estimation of IgG antibodies against pertussis toxin, filamentous hemagglutinin, pertactin, diphtheria toxoid, and tetanus toxoid. The MIA was calibrated using WHO reference standards and complied with US FDA, EMA, and ICH M10 guidelines. It showed good agreement with commercial ELISA assays and a positive correlation with toxin neutralization assays for pertussis and diphtheria.

The results showed that the vaccine was safe and well-tolerated in adults, with further clinical development recommended to evaluate its safety and immunogenicity. 23 subjects completed the study without any adverse events.
