In Situ Single-Molecule Imaging of MicroRNAs in Switchable Migrating Cells under Biomimetic Confinement

Summary: In this study, enzyme-free-mediated cascade amplified nanoprobes were designed for in situ single-molecule imaging of dual-microRNAs in switchable migrating cells. The differential expression and localization of dual-miRNAs were clearly exhibited in multiple cell lines due to the enhanced sensitivity via the cascade signal amplification strategy. Moreover, in situ three-dimensional imaging of dual-miRNAs in transition of cell migration phenotypes was successfully reconstructed in both non-confined and confined microenvironments in vitro, demonstrating differential spatial distribution in a single cell. This holds great promise for exploring the key roles of spatial RNA distribution at the single-molecule level in migrating cells, and for advancing the understanding of molecular mechanisms and physical principles in 3D cell migration in vivo.
