Summary: Adeno-associated virus (AAV) vectors are a promising delivery system for gene therapy. The current standard for AAV vector storage is deep-freezing below-60 degrees C, but there is a need for formulations that allow for storage at higher temperatures. This study evaluated different freeze-drying conditions for AAV8 vectors and found that stability can be improved by performing secondary drying at lower temperatures and maintaining residual moisture between 1.5 and 2%. Furthermore, certain formulations with specific salt concentrations and excipient compositions provided stable long-term storage at 2-8 degrees C and for at least 4 weeks at 25 degrees C.
