Summary: Selective functionalization of amines through cleavage of the C-N bond is challenging due to the high bond dissociation energy. Pre-activation of the C-N bond before cleavage has been developed as a strategy for overcoming this challenge. Conversion of amines into quaternary ammonium salts offers advantages such as easy preparation and stable storage. Recent studies have demonstrated the use of green electro- or photochemical tools for developing new transformations via oxidative addition of the C-N bond of quaternary ammonium salts.

Practical povidone iodine catalyzed transamidation from primary amides and amines

Summary: Non-harmful Povidone iodine (PVP-I) was used in the transamidation reaction to obtain N-substituted amides from various primary amides and amines. Our developed method successfully generated more than 45 N-substituted amides with excellent yields using diverse aromatic, heterocyclic, and aliphatic primary amides/amines. Drugs such as Melatonin and Efaproxiral were obtained with improved performance compared to previous reported results.

Organic photoredox catalyzed dealkylation/acylation of tertiary amines to access amides

Tert-Butyl Nitrite-initiated C-N Bond Cleavage of 1-Nitromethyl-N-aryltetrahydroisoquinolines: Synthesis of Furoxans with N-NO Skeleton

Summary: A series of furoxan derivatives with N-nitroso groups were efficiently synthesized by TBN-initiated radical sp(3) C-N bond cleavage of 1-nitromethyl-N-aryltetrahydroisoquinolines. This reaction introduces the biologically important furoxan skeleton and N-nitroso group into one molecule, significantly enhancing molecular complexity through a one-step transformation. Mechanistic studies reveal that this reaction is mediated by the in situ generated alpha-carbonyl nitrile oxide, which is formed via TBN-promoted C-N bond cleavage.

Summary: The challenging transamidation of unactivated tertiary amides has been accomplished via cooperative acid/iodide catalysis. This method provides a direct route to amide-to-amide bond interconversion with excellent chemoselectivity using equivalent amounts of amines. The combination of acid and iodide activates the amide C-N bond through electrophilic catalytic activation, enabling the production of new desired transamidated products with wide substrate scope.

Cobalt-Catalyzed Deaminative Amino- and Alkoxycarbonylation of Aryl Trialkylammonium Salts Promoted by Visible Light

Summary: In this study, we demonstrated an aminocarbonylation of aniline-derived trialkylammonium salts promoted by visible light with a simple cobalt catalyst. The reaction proceeds under mild conditions suitable for late-stage functionalization and is amenable to telescoped carbonylations directly from anilines. A range of alkylamines are successful partners, and alkoxycarbonylation is also demonstrated.

Electrochemical Oxidative C-C Bond Cleavage of Ketones for C-N Bond Formation: A Route to Amides

Summary: Herein, we report an efficient electrochemical method for the activation of the C-C bond of aryl ketones to prepare amides using aliphatic amines as the N source, without the need for catalysts or external oxidants. Under environmentally friendly electrolysis conditions, a series of amides were synthesized in good yield. Our control experiments demonstrated the significant role of electricity in this transformation.

Electrochemical Amidation: Benzoyl Hydrazine/Carbazate and Amine as Coupling Partners

Summary: An electrochemical reaction was utilized to couple benzoyl hydrazine/carbazate and primary/secondary amine, resulting in the cleavage and formation of C(sp(2))-N bonds. This method is metal-free and does not require an external oxidant, producing N-2 and H-2 as byproducts. Mechanistic studies discovered the in situ generation of acyl and N-centered radicals from benzoyl hydrazines and amines. The usefulness of this approach was demonstrated through the large-scale synthesis of bezafibrate, a hyperlipidemic drug.

Development of a Catalytic Ester Activation Protocol for the Efficient Formation of Amide Bonds using an Ar-I/HF•pyridine/mCPBA System

Summary: A new catalytic approach for the activation of esters, that possess a p-hydroxyphenyl group as a potential activating group on the ester residue, was developed. This approach involves the in-situ generation of a hypervalent iodine(III) species using an Ar-I/HF center dot pyridine/mCPBA system, which then leads to the smooth generation of a reactive acyl fluoride intermediate via the oxidative dearomatization of phenols. Various amides and peptides, including sterically hindered derivatives, can be efficiently synthesized using this method.

Electrooxidation-induced selective cleavage of C-N bonds of tertiary amines to access thioureas, selenoureas, and 2-aminated benzoselenazoles

Summary: A metal-free, operationally simple, and scalable electrooxidation method was reported for the selective cleavage of C-N bonds of tertiary amines, providing access to asymmetric thiourea, selenourea, and 2-aminated benzoselenazole derivatives. Over 50 examples were reported with yields greater than 96%. The readily accessible compounds 10c and 24c showed promising anticancer activity against mouse breast cancer and human lung adenocarcinoma cancer cell lines (A549), highlighting the importance of this methodology.

Cu-catalysed transamidation of unactivated aliphatic amides

In this study, we successfully achieved transamidation of unactivated aliphatic amides using a copper catalyst and chlorotrimethylsilane as an additive.
