Diversifying Amino Acids and Peptides via Deaminative Reductive Cross-Couplings Leveraging High-Throughput Experimentation

Summary: A deaminative reductive coupling method was developed for the efficient synthesis of noncanonical amino acids and diversification of peptides. The method allows the transformation of natural and commercially available amino acids to aryl alanines and derivatives with varying chain lengths. It has attractive features such as scalability, tolerance of pharma-relevant aryls and biorthogonal functional groups, and applicability to short and macrocyclic peptide substrates.

Reductive Cross-Coupling of Unreactive Electrophiles

Decarbonylative Synthesis of Aryl Nitriles from Aromatic Esters and Organocyanides by a Nickel Catalyst

Summary: A decarbonylative cyanation of aromatic esters with aminoacetonitriles in the presence of a nickel catalyst was developed using a thiophene-based diphosphine ligand, dcypt, to synthesize aryl nitrile without generating stoichiometric metal- or halogen-containing chemical wastes. A wide range of aromatic esters, including hetarenes and pharmaceutical molecules, can be converted into aryl nitriles.
