Targeting Xkr8 via nanoparticle-mediated in situ co-delivery of siRNA and chemotherapy drugs for cancer immunochemotherapy

Yuang Chen et al.

Summary: The activation of scramblases, a mechanism that regulates the exposure of phosphatidylserine on cell surfaces, plays an important role in tumor immunosuppression. In this study, the researchers found that chemotherapeutic agents induce overexpression of a specific scramblase, Xkr8, in cancer cells. They developed a nanocarrier that can deliver Xkr8 short interfering RNA and a cancer prodrug to tumors, leading to significant inhibition of tumor growth and increased antitumor immune response in animal models. Targeting Xkr8 in combination with chemotherapy may be a promising strategy for treating different types of cancers.

Free-Blockage Mesoporous Silica Nanoparticles Loaded with Cerium Oxide as ROS-Responsive and ROS-Scavenging Nanomedicine

Olha Purikova et al.

Summary: Mesoporous silica nanoparticles (MSNs) with a reactive oxygen species (ROS)-responsive nanogate have been extensively studied for biomedical applications. However, the current physical blockage methods used to control release are limited by their sealing ability and biocompatibility. In this study, researchers propose a design of free-blockage MSNs with methylthiopropyl units as ROS-responsive switches. By optimizing synthetic pathways, the quantity, localization, and chemical structure of the functional units, as well as the mesoporous structure of the silica, can be controlled to obtain desired products. The ROS-responsive methylthiopropyl groups can be oxidized to sulfoxides in the presence of H2O2, leading to the hydrophobic/hydrophilic conversion of the MSNs. As a proof-of-concept, ultrasmall cerium oxide nanoparticles are encapsulated into the functionalized MSNs and released within a short time period, effectively scavenging ROS. This study presents a novel design for a ROS-controlled release system, which can potentially be used in targeted drug delivery combined with antioxidant therapy.

Summary: The study compared the transcriptional profiles of human and mouse astrocytes, revealing species-specific responses to oxidative stress, hypoxic conditions, and inflammation. While gene expression in astrocytes showed extensive conservation between human and mouse samples, species-specific differences were observed in defense response and metabolism pathways. Human astrocytes were found to be more susceptible to oxidative stress than mouse astrocytes, with differences in mitochondrial physiology and detoxification pathways. Additionally, human and mouse astrocytes exhibited distinct molecular responses under hypoxic and inflammatory conditions.
