A ciprofloxacin derivative with four mechanisms of action overcomes paclitaxel resistance in p53-mutant and MDR1 gene-expressing type II human endometrial cancer

Suhaila O. Alhaj-Suliman et al.

Summary: Dysfunction of p53 gene and presence of MDR1 gene are associated with many malignant tumors, including endometrial cancer, and contribute to drug resistance. A new ciprofloxacin derivative, CIP2b, in combination with taxanes, showed synergistic cytotoxic effects against endometrial cancer cells with MDR1 expression and loss-of-function p53. CIP2b targets multiple pathways and enhances the effects of paclitaxel on microtubule assembly.

Engineering nanosystems to overcome barriers to cancer diagnosis and treatment

Suhaila O. Alhaj-Suliman et al.

Summary: In the past two decades, there has been increasing interdisciplinary research on the development of nanoparticles for medical applications. However, nanoparticles still face biological barriers and biodistribution challenges, limiting their overall clinical potential. To overcome these limitations, innovative modifications have been made to nanoparticle formulations designed for cancer imaging and treatment, aiming to enhance specificity and target diseased tissues. Recent advancements in technology and chemistry have been employed to modify the surface and characteristics of nanoparticles, in combination with identifying critical biomarkers, providing new strategies for improving nanoparticle specificity in cancer diagnosis and therapy. This review discusses the latest advances in nanoparticle design and engineering, and outlines future directions for the development of next-generation nanomedicines.

Light-activatable liposomes for repetitive on-demand drug release and immunopotentiation in hypoxic tumor therapy

Yuanyuan Yang et al.

Summary: A multifunctional light-activatable liposome has been engineered for repetitive on-demand drug release under discontinuous light irradiation, allowing for chemo-photodynamic therapy and immunopotentiation in hypoxic tumors. This system effectively remodels the redox balance in tumors, alleviating hypoxia and enhancing PDT efficacy, reversing cisplatin resistance, and polarizing tumor-associated macrophages to the immunocompetent M1 phenotype.

Self-targeting visualizable hyaluronate nanogel for synchronized intracellular release of doxorubicin and cisplatin in combating multidrug-resistant breast cancer

Wen Ma et al.

Summary: In this study, a self-targeting hyaluronate nanogel was developed for reversing drug resistance through synchronized pharmacokinetics, intratumoral distribution, and intracellular release of doxorubicin and cisplatin. This nanogel showed efficient drug delivery into drug-resistant breast cancer cells and enhanced antitumor activity, with potential for future clinical therapy of advanced cancers. Visualized synchronized drug delivery and imaging techniques facilitate the visualization of combination tumor chemotherapy.

Thiophene Derivative-Loaded Nanoparticles Mediate Anticancer Activity Through the Inhibition of Kinases and Microtubule Assembly

Somaya A. Abdel-Rahman et al.

Summary: Different tetrahydrobenzo[b]thiophene derivatives, especially the benzyl urea derivative BU17, have been identified as potent antitumor compounds with broad-spectrum activity against several cancer cell lines through inhibiting tubulin polymerization. BU17 induces apoptosis, potentially by targeting WEE1 kinase and tubulin. Furthermore, BU17-loaded PLGA nanoparticles exhibit significantly enhanced antitumor activity compared to the soluble form.
