Unraveling Presenilin 2 Functions in a Knockout Zebrafish Line to Shed Light into Alzheimer's Disease Pathogenesis

Lucia Barazzuol et al.

Summary: This study generated a zebrafish line with a psen2 knockout, and found that the absence of the protein does not significantly affect Notch signaling, but leads to an exaggeration of locomotor response, a decrease in ER-mitochondria contacts, and an increase in basal autophagy. The protein also plays a role in mitochondrial axonal transport, and its downregulation reduces organelle flux in zebrafish sensory neurons. These findings highlight zebrafish as a valuable model organism for studying PS2 functions and testing potential therapies for Alzheimer's disease.

High-throughput detection of craniofacial defects in fluorescent zebrafish

Joshua L. Everson et al.

Summary: Losses and malformations of cranial neural crest cell (cNCC) derivatives are a common feature of brain and face malformations. The etiology of these defects involves complex interactions between genetic and environmental factors. This study developed a high-throughput analysis method using zebrafish embryos to identify multifactorial interactions in the genesis of craniofacial defects. The results showed that exposure to environmental toxins and alcohol can reduce cNCC fluorescence, leading to craniofacial defects. The study also demonstrated the sensitivity of the assay in detecting alterations in Hedgehog signaling, a critical regulator of craniofacial development.

Syringol, a wildfire residual methoxyphenol causes cytotoxicity and teratogenicity in zebrafish model

P. Snega Priya et al.

Summary: This study evaluates the cytotoxic and teratogenic effects of methoxy phenols (MPs) generated by wildfire lignin on human embryonic kidney (HEK-293) cells and zebrafish embryos. The results indicate that MPs, particularly syringol, have toxic effects on both cells and embryos, including cell death, developmental abnormalities, and increased oxidative stress response. Therefore, syringol has the potential to pose serious health hazards in various living systems.

Astaxanthin mitigates oxidative stress caused by microplastics at the expense of reduced skin pigmentation in discus fish

Jun-Nan Huang et al.

Summary: This study aimed to determine whether astaxanthin could mitigate the oxidative stress caused by microplastics (MPs) but at the expense of reduced skin pigmentation in fish. Results showed that astaxanthin improved the oxidative stress caused by MPs, but reduced fish skin pigmentation.

Carboxin can induce cardiotoxicity in zebrafish embryos

Yong Huang et al.

Summary: Carboxin exhibits developmental and cardiotoxicity in zebrafish embryos, causing structural changes in the heart and disruption of heart developmental gene expression. Additionally, Carboxin induces oxidative stress and accumulation of reactive oxygen species (ROS) in zebrafish embryos, leading to mitochondrial damage and decreased ATPase activities. Carboxin also inhibits myocardial cell proliferation and decreases the number of blood cells in the heart, resulting in embryo death.

Knockdown of hspg2 is associated with abnormal mandibular joint formation and neural crest cell dysfunction in zebrafish

Barbara S. Castellanos et al.

Summary: Knockdown of HSPG2 in zebrafish resulted in abnormal development of the mandibular jaw joint, possibly due to dysfunction of neural crest cells. Early neural crest cell development was normal in morphant animals, but later stages showed a decrease in specific cell types in the mandibular jaw joint region. This suggests a complex role for HSPG2 in jaw joint formation and neural crest cell differentiation.

Survival and malformations rates, oxidative status in early life stages of Cyprinus carpio due to exposure to environmentally realistic concentrations of paracetamol

Veronica Margarita Gutierrez-Noya et al.

Summary: The study evaluated the toxic effects of paracetamol on early life stages of common carp, revealing that high concentrations of PCM resulted in reduced survival rate, changes in oxidative stress biomarkers, and developmental deformities. Carp embryos may serve as a useful model for assessing embryonic and teratogenic effects of drugs like PCM.

Diabetes, Oxidative Stress, and DNA Damage Modulate Cranial Neural Crest Cell Development and the Phenotype Variability of Craniofacial Disorders

Sharien Fitriasari et al.

Summary: Craniofacial malformations are common birth defects in humans and have significant functional, aesthetic, and social consequences. Over 700 distinct disorders have been described, but the genetic, environmental, and developmental origins remain unclear. Phenotypic variation seems to be a result of both genetic and non-genetic factors, with environmental factors such as maternal diabetes playing a role in increasing the risk of craniofacial birth defects.
