Summary: This study investigated the role of stress-induced phosphoprotein 1 (STIP1), heat shock protein (HSP) 70, and HSP90 in the ubiquitination of connexin 43 (Cx43) in rat H9c2 cardiomyocytes. The results showed that STIP1 bound to HSP70 and HSP90, while Cx43 bound to HSP40, HSP70, and HSP90. Overexpression of STIP1 facilitated the transition of Cx43-HSP70 to Cx43-HSP90 and inhibited Cx43 ubiquitination, while knockdown of STIP1 had the opposite effects. Inhibition of HSP90 counteracted the inhibitory effect of STIP1 overexpression on Cx43 ubiquitination. STIP1 is found to suppress Cx43 ubiquitination in H9c2 cardiomyocytes by promoting the transition of Cx43-HSP70 to Cx43-HSP90.

Influence of the exposure concentration of dissolved cadmium on its organotropism, toxicokinetic and fate in Gammarus fossarum

Summary: This study aimed to investigate the effects of Cd concentration on organotropism, toxico-kinetics, and fate of Cd in gammarids. The results showed that Cd concentration did not significantly modulate the bioconcentration capacity and Cd distribution in different organs. However, Cd accumulation rates in organs (except intestines) were dependent on the exposure concentration. The in vivo management of Cd among organs remained qualitatively unchanged according to exposure concentration. This study highlights the importance of implementing toxico-kinetic approaches to enhance the understanding of the toxicity risk of contaminants in invertebrates.

StatsPro: Systematic integration and evaluation of statistical approaches for detecting differential expression in label-free quantitative proteomics

Summary: Quantitative label-free mass spectrometry is a powerful technology for analyzing proteins in complex biological samples and detecting differentially expressed proteins (DEPs) under different experimental conditions. Despite the existence of numerous statistical methods for DEP detection in proteomics, it remains a challenge for many scientists to choose the appropriate procedure. A new web tool, StatsPro, has been developed to provide a systematic approach for evaluating and implementing different statistical methods for proteomics research, offering convenience and support for method selection.

Improving the proteome coverage of Daphnia magna - implications for future ecotoxicoproteomics studies

Summary: Aquatic pollution is a growing problem that requires extensive research to understand its consequences and find solutions. The crustacean Daphnia is a key species in lake ecosystems, making it suitable for studying the effects of freshwater pollution. Proteome analysis of Daphnia is hindered by highly abundant proteins, but a new method based on LC-MS/MS analysis of whole daphnids and dissected Daphnia gut was established to improve proteome coverage. Using this method, researchers identified a large number of protein groups in Daphnia and Daphnia gut samples, demonstrating the effectiveness of the approach.

Organ-specific accumulation of cadmium and zinc in Gammarus fossarum exposed to environmentally relevant metal concentrations*

Summary: One of the best approaches for improving the assessment of metal toxicity in aquatic organisms is to study their organotropism through kinetic experiments of metal bioaccumulation. This study compared the organ-specific metal accumulation of non-essential (Cd) and essential (Zn) metals in Gammarus fossarum. The results showed different distribution patterns of Cd and Zn concentrations, slower elimination of Cd, and a major role of intestines in the uptake of waterborne Cd and Zn.

Summary: Silver nanomaterials are widely used and studied, and in this study, the differential impact of AgNM300K and AgNO3 on protein and metabolite expression in Enchytraeus crypticus was assessed using high-throughput techniques. Results showed that the two materials elicited different responses in biological pathways, with AgNO3 causing a dose response increase of proteins/metabolites and NM300K causing more upregulation at lower concentrations. This study contributes to understanding the molecular mechanisms underlying the effects of silver nanomaterials on organisms.

Summary: This research evaluated the antioxidant capability of quercetin against silver nanoparticles toxicity towards the lipid contents of ovarian, nervous, and hepatic systems as well as skeletal muscles. The findings suggest that unsaturated acyl chains, particularly HUFAs, in the liver tissue and oocyte cell are highly susceptible to peroxidation or degeneration by AgNPs. The alteration in HUFAs and PUFAs of the liver and oocyte could impact on maternal and offspring health and consequently alter long-term population dynamics of aquatic animals.
